大鼠青光眼滤过泡瘢痕化模型中MMP-2及TIMP-2表达的实验研究

【摘要】:研究目的:建立大鼠青光眼结膜滤过泡瘢痕化模型:观察结膜下组织瘢痕化过程中基质金属蛋白酶-2(Matrix metalloproteinase-2, MMP-2)及其组织金属蛋白酶抑制物一2(tissue inhibitors of Metalloproteinase-2, TIMP-2)的表达情况,初步探讨MMP-2及TIMP-2在瘢痕化形成中的作用。 方法:选用健康成年SD大鼠60只,通过前房引流管植入术,成功建立青光眼滤过泡瘢痕化模型。术后观察滤过泡的形成情况,并分别于术后1d、3d、5d、7d、14d、28d取结膜滤过区组织分别进行HE染色和western blot检测,观察各时间点MMP一2及TIMP-2的表达情况。 结果:1.所有术眼在术后第1天均能形成不同程度隆起明显的滤过泡。滤过泡维持时间为7-17天,平均生存时间是12天;2.HE染色发现:滤过泡瘢痕形成的过程中可见炎症细胞、成纤维细胞、血管形成及胶原纤维形成等一系列动态变化:3.半定量的western blot统计学结果显示:MMP-2在术后第1天表达降低,术后第3天增高,持续增高至术后第7天达峰值,随后逐渐降低至术后第28天,但仍高于对照组,差异具有显著统计学意义(P0.01)。TIMP-2在术后第1天开始增高,持续增高至术后第5天达峰值,随后逐渐降低至术后第28天,但仍高于对照组,差异具有显著统计学意义(P0.01)。 结论:在大鼠结膜下组织创伤修复过程中,MMP-2及TIMP-2在结膜下组织中表达量发生变化,说明二者参与结膜滤过泡瘢痕化修复过程。提示研究TIMP-2对MMP-2的特异性抑制作用,可能为开发新的抗瘢痕化药物提供依据。
[Abstract]:Objective: to establish a rat model of glaucoma and conjunctival bleb scarring: to observe the expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in subconjunctival tissue scarring, and to observe the expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2). To explore the role of MMP-2 and TIMP-2 in scarring formation. Methods: 60 healthy adult SD rats were used to establish the model of glaucoma bleb scarring successfully by anterior chamber drainage tube implantation. The formation of filtering bleb was observed after operation. He staining and western blot detection were performed on the conjunctival filtration area at 1 day, 3 days, 5 days, 7 days, 14 days and 28 days, respectively. The expression of MMP-2 and TIMP-2 were observed at each time point. Results 1. On the first day after operation, all operative eyes could form obvious blebs with different degrees of bulge. The mean survival time was 12 days and the mean survival time was 12 days. 2. The inflammatory cells were found in the process of bleb scar formation. A series of dynamic changes, such as fibroblast, angiogenesis and collagen fiber formation, showed that the expression of western blot decreased on the first day after operation, increased on the third day after operation, and continued to increase to the peak value on the 7th day after operation. Then decreased gradually to 28 days after operation, but still higher than that of the control group. The difference was statistically significant (P 0.01). TIMP-2 began to increase on the first day after operation, continued to increase to the peak value on the 5th day after operation, and then gradually decreased to the 28th day after operation. But still higher than the control group, the difference was statistically significant (P 0.01). Conclusion: the changes of expression of MMP-2 and TIMP-2 in subconjunctival tissue during the process of subconjunctival wound repair in rats indicate that they are involved in the scarring repair process of conjunctival filtration bleb. It is suggested that the specific inhibitory effect of TIMP-2 on MMP-2 may be studied. It may provide evidence for the development of new anti-scar drugs.
