MIF和IL-5在鼻息肉组织中的表达及意义

【摘要】:目的:通过检测鼻息肉组织中巨噬细胞移动抑制因子(MIF)、白细胞介素-5(IL-5)的表达和嗜酸性粒细胞(Eos)的浸润情况,并采用视觉模拟量表(VAS)和Lund-Mackay评分法评估鼻息肉患者病情严重程度,分析MIF、IL-5表达与Eos浸润程度间的相关性及两因子表达与疾病严重程度间的关系,探讨MIF和IL-5在鼻息肉发生过程中的作用,并分析其可能作用途径。方法:采用VAS评分和Lund-Mackay评分评估鼻息肉患者病情严重程度;应用HE染色(苏木精-伊红染色法)检测鼻息肉组织、正常中鼻甲粘膜组织中EOS的浸润程度并计数;采用免疫组织化学法(SP法)检测鼻息肉组织、正常中鼻甲组织中MIF、IL-5的表达并进行计数,分析二者与疾病严重程度及EOS浸润程度的相关性。结果:1.EOS在鼻息肉组织中的浸润程度(39.30±4.49)个/HP明显高于在对照组中的浸润数量(7.70±3.19)个/HP,p0.05;2.MIF在鼻息肉组织中的表达(0.3693±0.0535)明显高于对照组中的表达量(0.1557±0.0395),p0.05;3.IL-5在鼻息肉中的表达量(0.7378±0.2262)明显高于在对照组中的表达量(0.2253±0.1061),p0.05;4.MIF、IL-5表达与Eos浸润程度间呈正向相关性(r分别为0.793、0.843,p均0.05);5.鼻息肉患者VAS评分与MIF、IL-5表达量间呈正相关性(r分别为0.598、0.769,p均0.05),鼻窦CT Lund-Mackay评分与二者表达同样呈正相关性(r分别为0.613、0.626,p均0.05)。结论:1.MIF、IL-5在鼻息肉组织中的高表达,说明二者共同参与了鼻息肉的发生发展;2.MIF、IL-5表达与EOS浸润程度间呈正相关,说明二者可能通过增加EOS浸润程度促使鼻息肉的形成;3.MIF、IL-5表达与患者病情严重程度间呈正相关,表明二者的表达可在一定程度上评估患者疾病的严重程度。
[Abstract]:Objective: to detect the expression of macrophage migration inhibitory factor (MIFN) and interleukin-5 (IL-5) and the infiltration of eosinophil in nasal polyps, and to evaluate the severity of nasal polyps by visual analogue scale (VAS) and Lund-Mackay score. To analyze the relationship between the expression of IL-5 and the degree of Eos invasion, and the relationship between the expression of two factors and the severity of the disease, and to explore the role of MIF and IL-5 in the pathogenesis of nasal polyps. Methods: VAS and Lund-Mackay scores were used to evaluate the severity of nasal polyps, and HE staining (hematoxylin eosin staining) was used to detect nasal polyps. The infiltration degree and count of EOS in normal mucosa of middle turbinate, the expression of MIFIL-5 in nasal polyp tissue and normal middle turbinate tissue were detected and counted by immunohistochemical method. Results 1. The infiltration degree of EOS in nasal polyps was 39.30 卤4.49) / HP was significantly higher than that in control group (7.70 卤3.19) and the expression of EOS in nasal polyps was 0.3693 卤0.0535). The expression of IL-5 in nasal polyps was 0.7378 卤0.2262), which was significantly higher than that in control group (0.2253 卤0.1061). The positive correlation between the expression of IL-5 and the degree of Eos infiltration was 0.7930.843 and 0.05 respectively. The positive correlation between the VAS score and the expression of IL-5 in nasal polyps was 0.05% and 0.2253 卤0.1061 respectively, and the positive correlation between the expression of IL-5 and the degree of Eos infiltration was 0.7930.843, P = 0.05, respectively. The positive correlation between the expression of IL-5 in nasal polyps and the expression of IL-5 in nasal polyps was significantly higher than that in the control group (0.2253 卤0.1061). There was a positive correlation between 0. 5988 and 0. 769 (p = 0. 05), and there was a positive correlation between CT Lund-Mackay score of paranasal sinus and the expression of both. Conclusion: 1. MIF- IL-5 is highly expressed in nasal polyps, and the positive correlation is 0. 613 ~ 0. 626p, P = 0. 05 respectively. 2. There was a positive correlation between the expression of IL-5 in nasal polyps and the degree of EOS infiltration. It was suggested that the expression of IL-5 in nasal polyps was positively correlated with the severity of the disease by increasing the degree of EOS infiltration. These results suggest that the expression of both can be used to evaluate the severity of the disease to some extent.
