Phosducin基因在糖尿病大鼠模型视网膜病变过程中表达变化研究

【摘要】:目的: 检测Phosducin基因在不同病程糖尿病大鼠模型视网膜组织上的表达及定位,探讨其在糖尿病大鼠视网膜病变过程中表达变化及作用,为进一步深入研究糖尿病视网膜病变的发病机制提供实验依据。 方法: 选取实验组糖尿病GK大鼠32只和对照组Wistar大鼠32只,按糖尿病病程分为四组,即糖尿病4W组、8W组、16W组、24W组,每组8只。对照组与实验组周龄相对应分组,每组8只。采用荧光素眼底血管造影(fundus fluorescein angiography, FFA)技术,分析大鼠模型视网膜病变程度与病程的关系。采用实时荧光定量多聚酶链反应(RT-real-time PCR)技术,分析Phosducin基因在不同病程糖尿病大鼠视网膜组织上的表达量,并应用免疫组化技术检测Phosducin基因在糖尿病大鼠视网膜组织上的表达定位。 结果: 1、FFA检查结果显示,GK大鼠DR的发生并不完全与糖尿病病程及血糖高低成正相关。 2、RT-PCR结果显示,Phosducin在对照组大鼠各期视网膜的表达变化无明显差异(P0.05)。与对照组相比,糖尿病组各期视网膜其表达是下降的,且随着周龄的增加,表达越来越低,即4W时开始降低,24W时降到最低(P0.01)。 3、免疫组化结果显示Phosducin表达在视椎视杆层和外核层细胞,阳性细胞在对照组大鼠各期无明显差异(P0.05)。与对照组相比,糖尿病组各期视网膜其阳性表达是下降的,且随着周龄的增加,表达逐渐降低,以内核层变化最为明显,即4W时开始降低,24W时降到最低(P0.01)。 结论: 1、糖尿病GK大鼠DR的发生并不完全与糖尿病病程及血糖高低成正相关。 2、Phosducin基因是视觉适应的重要调控因子,在视觉传导通路中起重要的调节作用,可能与神经元细胞功能障碍有关,其表达与糖尿病的病程密切相关。 3、Phosducin基因表达异常变化,可能是糖尿病早期视功能障碍的原因之一。
[Abstract]:Objective:To detect the expression and localization of Phosducin gene in the retina of diabetic rats with different course of disease, and to explore the changes and roles of Phosducin gene expression in the process of diabetic retinopathy.To provide experimental evidence for further study of the pathogenesis of diabetic retinopathy.Methods:Thirty-two diabetic GK rats in the experimental group and 32 Wistar rats in the control group were divided into four groups according to the course of diabetes.The control group and the experimental group were divided into groups of 8 rats in each group.Fundus fluorescein angiographic (FFAA) technique was used to analyze the relationship between the degree of retinopathy and the course of retinopathy in rats with fundus fluorescein angiography (FFA).The expression of Phosducin gene in the retina of diabetic rats with different course of disease was analyzed by real-time real-time PCR and the localization of Phosducin gene expression in the retina of diabetic rats was detected by immunohistochemical technique.Results:The results of FFA showed that Dr in GK rats was not positively correlated with the course of diabetes and the level of blood sugar.2the results of RT-PCR showed that there was no significant difference in the expression of Phosducin in the retina of rats in the control group.Compared with the control group, the expression of retina decreased in each stage of diabetic group, and with the increase of week age, the expression became lower and lower, that is, at 4 W, the expression began to decrease to the lowest level (P 0.01) at 24 W.3Immunohistochemical results showed that Phosducin was expressed in the cells of the optic rod layer and the outer nuclear layer of the optic vertebrae, and there was no significant difference in the positive cells between the control group and the control group (P 0.05).Conclusion:1. The incidence of Dr in diabetic GK rats was not positively correlated with the course of diabetes and the level of blood glucose.2 Phosducin gene is an important regulatory factor for visual adaptation, which plays an important regulatory role in the visual conduction pathway and may be related to neuronal dysfunction, and its expression is closely related to the course of diabetes mellitus.3 abnormal expression of Phosducin gene may be one of the causes of visual dysfunction in the early stage of diabetes mellitus.
