喉癌中钙网织蛋白和CD47的表达及其与T淋巴细胞浸润的相关性

【摘要】:目的:通过检测喉鳞状上皮细胞癌(laryngeal squamous cell carcinoma,LSCC)组织中促吞噬信号作用的钙网织蛋白(calreticulin,CRT)和抗吞噬信号作用CD47的表达,探讨二者在LSCC发生发展中的作用及其与T淋巴细胞浸润的相关性。方法:应用逆转录酶-聚合酶链反应(RT-PCR)检测42例LSCC组织及18例癌旁正常鳞状上皮组织中CRT和CD47mRNA的表达水平,免疫组化S-P法检测68例原发性LSCC及20例癌旁正常组织中CRT和CD47蛋白的表达情况,并用CD3及CD45RO单克隆抗体标记癌组织实质及间质中T淋巴细胞浸润情况。结果:在LSCC组织中,CRT和CD47mRNA和蛋白的表达均高于癌旁正常鳞状上皮组织(P0.05);CRT及CD47蛋白随着LSCC的肿瘤T分期、淋巴结转移及临床分期的增加其表达增加(P0.05),CD47与CRT蛋白的表达有一种较强的正相关性(r=0.89P0.05);CRT蛋白的表达与CD45RO+T淋巴细胞的高浸润相关(P0.05),CD47蛋白的表达与CD3+T淋巴细胞及癌组织间质中CD45RO+T淋巴细胞的浸润无相关性(P0.05). 结论:CRT和CD47在LSCC的发生、发展过程中可起重要作用,,CRT的表达与CD45RO+T淋巴细胞的高浸润相关,可能是喉鳞癌的一种新的抗肿瘤免疫机制。
[Abstract]:Objective: to detect the expression of calreticulinus calreticulinus (CRT) and CD47 in laryngeal squamous cell carcinoma of larynx squamous cell carcinoma (LSCC).To explore the role of both of them in the pathogenesis and development of LSCC and the correlation between them and T lymphocyte infiltration.Methods: reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of CRT and CD47mRNA in 42 cases of LSCC and 18 cases of adjacent normal squamous epithelium.Immunohistochemical S-P method was used to detect the expression of CRT and CD47 protein in 68 cases of primary LSCC and 20 cases of adjacent normal tissues. T-lymphocyte infiltration in parenchyma and interstitial tissue was labeled with CD3 and CD45RO monoclonal antibodies.Results: the expression of CRT, CD47mRNA and protein in LSCC was higher than that in normal squamous epithelium adjacent to the carcinoma (P0.05) and CD47 protein was associated with the T stage of LSCC.The expression of CD47 and CRT protein in lymph node metastasis and clinical stage increased. There was a strong positive correlation between the expression of CD47 and CRT protein and the high infiltration of CD45RO T lymphocytes. The expression of CD47 protein was correlated with the expression of CD3 T lymphocytes and carcinoma.There was no correlation between the infiltration of CD45RO T lymphocytes in the interstitial tissue (P 0.05).Conclusion the expression of LSCC and CD47 may play an important role in the pathogenesis and development of LSCC. The expression of CRT may be associated with the high infiltration of CD45RO T lymphocytes, which may be a new anti-tumor immune mechanism of laryngeal squamous cell carcinoma.
