AMPA受体在CRS嗅觉障碍大鼠嗅球中的表达变化

【摘要】:目的:观察AMPA受体(alpha-Amino-3-hydroxy-5-methyl-4-isoxazole-propionic Acid,α-氨基羟甲基恶唑丙酸)在慢性鼻及鼻窦炎(chronic rhinosinusitis,CRS)伴嗅觉障碍大鼠嗅球中的表达变化,探讨CRS伴嗅觉障碍的发病机制。 方法:采用鼻腔阻塞加肺炎链球菌接种的方法建立SD大鼠CRS伴嗅觉障碍模型。HE染色观察嗅粘膜形态变化,食物小球埋藏法(buffed food pellet test,BFPT)评估大鼠嗅觉功能, RT-PCR及免疫荧光两种方法检测嗅球中AMPA受体的表达。 结果:对照组、造模4周组、8周组及12周组嗅粘膜出现急性到慢性的变化过程,包括粘膜层增厚,,腺体增加,中性粒细胞、浆细胞、淋巴细胞增加。在慢性炎症过程中,淋巴细胞是主要浸润细胞。BFPT检测各模型组与对照组比较有差异,12周组出现明显的嗅觉障碍(P0.01)。12周模型组中嗅球GluR2mRNA的表达较正常组明显下降(P0.05)。免疫荧光检测嗅球GluR2表达在12周组荧光强度弱于前3组。与PCR结果一致。 结论:采用鼻腔阻塞加肺炎链球菌接种的方法可以成功建立CRS嗅觉障碍的大鼠模型。AMPA受体的表达下降可能参与CRS引起嗅觉障碍的发病机制。
[Abstract]:Objective: To observe the changes in the expression of AMPA receptor (alpha-Amino-3-hydroxy-5-methyl-4-isoxazole-propionic Acid, alpha aminomethyloxaloxalpropionic acid) in the olfactory bulb of rats with chronic rhinosinusitis (chronic rhinosinusitis, CRS) with olfactory disorder, and to explore the pathogenesis of the olfactory disorder in CRS.
Methods: using the method of nasal obstruction and Streptococcus pneumoniae inoculation, the morphologic changes of olfactory mucosa were observed by.HE staining of CRS with olfactory disorder model in SD rats. The olfactory function of rats was evaluated by buffed food pellet test (BFPT), and the expression of AMPA receptors in the olfactory bulb was detected by two methods of RT-PCR and immunofluorescence.
Results: in the control group, the olfactory mucosa of the 4 weeks group, the 8 week group and the 12 week group appeared acute to chronic changes, including the thickening of the mucous membrane, the increase of the gland, the increase of the neutrophils, plasma cells and lymphocytes. In the process of chronic inflammation, the lymphocyte was the main infiltrating cell.BFPT in each model group compared with the control group, and in the 12 week group. The expression of olfactory bulb GluR2mRNA in the.12 week model group of P0.01 was obviously lower than that in the normal group (P0.05). The fluorescence intensity of the olfactory bulb GluR2 expression in the 12 week group was weaker than the first 3 groups, and the results were consistent with the results of PCR.
Conclusion: using nasal obstruction and Streptococcus pneumoniae inoculation, the decrease of.AMPA receptor expression in rat model of CRS olfactory disorder may be involved in the pathogenesis of CRS induced olfactory disorder.
