DMBT1基因在鼻息肉发生发展中的作用及其机制研究

【摘要】:目的探讨DMBT1基因在鼻息肉发生发展中的作用及其机制。方法用shRNA敲降法建立体外稳定低表达DMBT1的鼻黏膜上皮细胞系,实验分三组:实验组(shRNA敲降DMBT1的鼻黏膜上皮细胞),对照组(未做任何处理的鼻黏膜上皮细胞),空载体组(转染空载体的鼻黏膜上皮细胞),CCK8法及流式细胞术检测各组的鼻黏膜上皮细胞的增殖、凋亡及细胞周期变化,western-blot检测各组的鼻黏膜上皮细胞中AKT、P53、STAT3及ERK1/2表达变化,并观察DMBT1作用的信号通路。结果实验组与对照组比较:鼻黏膜上皮细胞增殖增加,凋亡减少,细胞周期中G1期细胞明显减少,G2和S期细胞明显增多;AKT明显上调(P㩳0.05),P53蛋白明显下调(P㩳0.05);ERK1/2、STAT3的表达虽有微小差异,但差异无统计学意义(P㧐0.05)。对照组与空载体组比较:细胞增殖凋亡及细胞周期改变和AKT、P53、ERK1/2、STAT3差异均无统计学意义(P㧐0.05)。结论DMBT1基因在鼻息肉发生发展中起重要作用,其机制可能为:DMBT1表达下调通过“AKT-P53”信号通路促使鼻黏膜上皮细胞增殖增加、凋亡减少,促进鼻息肉产生。
[Abstract]:Objective to investigate the role and mechanism of DMBT1 gene in the development of nasal polyps. Methods nasal mucosal epithelial cell lines with stable and low expression of DMBT1 were established by shRNA knockdown in vitro. The experiment was divided into three groups: the experimental group was used to knock down the nasal mucosal epithelial cells of DMBT1, the control group was used to detect the nasal mucosal epithelial cells without any treatment, and the empty vector group (the empty vector transfected nasal mucosal epithelial cells was detected by CCK8 method and flow cytometry). The proliferation of nasal mucosal epithelial cells, Apoptosis and cell cycle changes were detected by Western blot in nasal epithelial cells of each group. The expression of AKTP-53-STAT3 and ERK1/2 were detected and the signal pathway induced by DMBT1 was observed. Results compared with the control group, the proliferation and apoptosis of nasal epithelial cells in the experimental group were increased, and the number of cells in G _ 1 phase and S phase in the cell cycle were significantly decreased. AKT significantly up-regulated the expression of P0. 05 and P0. 05 and ERK1 / 2 + STAT3, although there was a slight difference. But the difference was not statistically significant. There was no significant difference between control group and empty carrier group in cell proliferation, apoptosis, cell cycle change and AKT53ERK1 / 2STAT3. Conclusion DMBT1 gene plays an important role in the occurrence and development of nasal polyps, and its mechanism may be that the down-regulation of the expression of DMBT1 may promote the proliferation of nasal epithelial cells and decrease of apoptosis through the "AKT-P53" signaling pathway, thus promoting the production of nasal polyps.
