年龄相关性白内障患者晶状体前囊膜20s蛋白酶体α2亚基的表达研究

【摘要】:目的:通过对不同混浊程度及不同核硬度的年龄相关性白内障(Age-related cataract,ARC)患者的晶状体前囊膜20s蛋白酶体α2亚基的蛋白表达进行检测分析,以期了解不同混浊程度及不同核硬度的白内障患者其晶状体老化与20s蛋白酶体α2亚基的蛋白表达变化之间的关系及意义。方法:收集行超声乳化或小切口白内障摘除术的前囊膜120例(核性60例,皮质性60例)。根据晶状体核的硬度及混浊程度的不同进行分组,分为核性组(60例60眼)和皮质组(60例60眼)其中核性组按Emery分级标准分为两组,即A组:年轻核(I-II度),B组:老核(IV-V度);皮质组按LOCSII标准分为两组,即A组:轻度混浊(C0-C2),B组:重度混浊(C4-C5)。应用Western-Blotting法检测20s蛋白酶体α2亚基在核性组和皮质组ARC患者晶状体前囊膜中的蛋白表达水平。结果:核性组白内障中,A组中20s蛋白酶体α2亚基蛋白表达较B组增强((0.99±0,02)VS(0.58±0.08)),差异有统计学意义(P0.01);皮质组白内障中,A组中20s蛋白酶体α2亚基蛋白表达较B组增强((1.26±0,15)VS(0.89±0.09)),差异有统计学意义(P0.05);也即表示核性白内障中年轻核比老核20s蛋白酶体α2亚基的蛋白表达含量高,皮质白内障中轻度混浊白内障比重度混浊白内障20s蛋白酶体α2亚基的蛋白表达含量高。结论:ARC患者前囊膜中20s蛋白酶体α2亚基的蛋白表达含量随晶状体混浊程度及核的硬度加深而降低。
[Abstract]:Objective: to study the expression of proteasome 伪 2 subunit in anterior capsule of lens in patients with age-related cataract (Age-related cataract) with different degree of opacity and nuclear hardness. The purpose of this study was to investigate the relationship between lens aging and proteasome 伪 2 subunit protein expression in cataract patients with different opacity and nuclear hardness. Methods: 120 cases of anterior capsule (60 nuclear and 60 cortical) underwent phacoemulsification or small incision cataract extraction. According to the hardness of lens nucleus and the degree of opacity, they were divided into two groups: nuclear group (60 cases, 60 eyes) and cortical group (60 cases, 60 eyes). The nuclear group was divided into two groups according to Emery grading standard. Group A was divided into two groups according to LOCSII standard: group A: young nucleus: group B: IV-V degree of old nucleus, group A: mild turbidity: group C 0-C 2: group B: severe opacification: C4-C5. The expression of 伪 2 subunit of 20 s proteasome in anterior capsule of lens of ARC patients in nuclear and cortical groups was detected by Western-Blotting method. Results: the expression of proteasome 伪 2 subunit protein in group A was significantly higher than that in group B (0.99 卤0 02VSN 0.58 卤0.08), and that in group A was higher than that in group B (1.26 卤0 015VSU 0.89 卤0.09), and the expression of proteasome 伪 2 subunit in group A was significantly higher than that in group B (P < 0. 01), and the expression of proteasome 伪 2 subunit in group A was significantly higher than that in group B (P < 0. 01). The protein expression of 伪 2 subunit in the young nucleus is higher than that in the old nucleus, which means that the protein expression of 伪 2 subunit in the young nucleus is higher than that in the old nucleus. The protein expression of proteasome 伪 2 subunit in mild cataract was higher than that in severe cataract. Conclusion the protein expression of proteasome 伪 2 subunit in the anterior capsule of the patients with 1% ARC decreased with the degree of lens opacity and the hardness of nucleus.
