麦考酚酸对翼状胬肉成纤维细胞抑制作用的实验研究

【摘要】:目的研究麦考酚酸(MPA)对翼状胬肉组织成纤维细胞(PFB)活性的影响。方法体外培养PFB及Tenon囊成纤维细胞(TFB),以丝裂霉素C(MMC)为对照,MTT比色法、5-溴脱氧尿嘧啶(5-bromo-2-deoxyuridine,Brdu)掺入法观察MPA对PFB的生长抑制作用,以流式细胞术等检测MPA对PFB、TFB的细胞毒性作用、细胞周期及凋亡的影响。结果 MPA可显著抑制PFB、TFB的增殖,呈时间、剂量依赖性,且不引起TFB的凋亡或坏死。MPA作用后,PFB的G0/G1期比例增加,S期及G2/M期比例减少。结论 MPA对PFB有显著的抑制作用,但不引起TFB的凋亡或坏死,且主要抑制在PFB的DNA合成期。
[Abstract]:Objective to study the effect of mycophenolic acid (MPA) on PFB activity in pterygium tissue fibroblasts. Methods PFB and Tenon capsule fibroblasts were cultured in vitro, and mitomycin C MMC was used as control. The inhibitory effect of MPA on the growth of PFB was observed by 5-bromo-2-deoxyuridine incorporation, and the cytotoxicity of MPA to PFB was detected by flow cytometry. Cell cycle and apoptosis. Results MPA could significantly inhibit the proliferation of TFB in a time-and dose-dependent manner, and did not induce apoptosis or necrosis of TFB. After treatment, the proportion of G0/G1 phase of PFB increased and the proportion of G _ 2 / M phase decreased. Conclusion MPA has a significant inhibitory effect on PFB, but does not induce apoptosis or necrosis of TFB, and is mainly inhibited in the DNA synthesis phase of PFB.
