辛伐他汀对视神经夹伤后小鼠视网膜神经节细胞的保护作用

【摘要】:目的探讨辛伐他汀对视神经损伤后(optic nerve crush,ONC)视网膜神经节细胞(retinal ganglion cell,RGC)的保护作用。方法将60只昆明鼠分为正常组、假损伤组、损伤组和辛伐他汀修复组。正常组不作任何处理,假损伤组只暴露视神经不损伤,损伤组行左眼视神经夹伤手术后,玻璃体内注射平衡溶剂(50 mg·m L~(-1)乙醇和1 mol·L~(-1)Na OH的混合溶剂,并用1 mol·L~(-1)盐酸激活并调节p H值为7.2),辛伐他汀修复组行左眼视神经夹伤手术后,玻璃体腔注射不同浓度辛伐他汀(0.5g·L~(-1)、1.0 g·L~(-1)、1.5 g·L~(-1))。Brn3a免疫荧光染色、HE染色、甲苯胺蓝染色检测RGC凋亡及视神经的病理变化。结果术后7 d,损伤组RGC凋亡明显,RGC存活率降低至(35.1±3.9)%,RGC层到外核层厚度由(123.13±1.04)μm降低至(97.48±2.33)μm,与正常组相比,差异有统计学意义(P0.01);视神经纤维束状排列消失,神经胶质细胞轴突聚集成团,肿胀变形,退行性病变严重,与正常组相比差异显著。玻璃体内注射1.0 g·L~(-1)辛伐他汀后,RGC存活率增加至(76.3±3.7)%(P0.05),RGC层到外核层厚度增加至(111.39±4.06)μm,与损伤组相比,差异有统计学意义(P0.01),视神经退行性病变明显改善,胶质细胞轴突和神经纤维束状结构趋于正常。结论玻璃体内注射辛伐他汀可以减缓视神经退行性病变,提高RGC存活率。
[Abstract]:Objective to investigate the protective effect of simvastatin on retinal ganglion cells (RGCs) of retinal ganglion cells (RGCs) after optic nerve injury. Methods 60 Kunming rats were divided into normal group, sham injury group, injury group and simvastatin repair group. No treatment was given in the normal group, but only the optic nerve was exposed in the sham injury group. After the operation on the left eye optic nerve, the mixture of ethanol and 1 mol L~(-1)Na OH was injected intravitreally into the vitreous. The left optic nerve injury was performed in the simvastatin repair group. 0.5 g / L simvastatin was injected into the vitreous body and 1.0 g / L of simvastatin was injected into the vitreous cavity and the pH value was adjusted to 7.2g / h. He staining was performed with different concentrations of simvastatin (0.5 g / L) / L ~ (-1) / L ~ (-1) and 1.0 g / L ~ (+) ~ (-1) / L ~ (-1) / L ~ (-1) / L ~ (-1) ~ (-1). The apoptosis of RGC and the pathological changes of optic nerve were detected by toluidine blue staining. Results on the 7th day after operation, the survival rate of RGC decreased to 35.1 卤3.9 渭 m from 123.13 卤1.04 渭 m to 97.48 卤2.33 渭 m in the injured group. Swelling, deformability and degenerative lesion were serious, and the difference was significant compared with the normal group. The survival rate of RGC increased to 76.3 卤3.7m after intravitreous injection of 1.0 g / L ~ (-1) simvastatin, and the thickness of RGC layer to outer nuclear layer increased to 111.39 卤4.06 渭 m. Compared with the injured group, the difference was statistically significant (P 0.01), and the optic neurodegenerative lesion was significantly improved. Glial axons and nerve fiber bundles tend to be normal. Conclusion Intravitreous injection of simvastatin can reduce the degeneration of optic nerve and improve the survival rate of RGC.
