MiR-148a在鼻咽癌中的表达及其对肿瘤细胞生物学功能的影响

【摘要】:目的:探讨MiR-148a在鼻咽癌细胞中的表达水平变化及其对鼻咽癌肿瘤细胞生物学功能的影响。方法:采用Real-time PCR方法检测MiR-148a在鼻咽癌细胞系CNE2中的表达水平变化,并通过MTT实验、克隆形成实验和流式细胞术检测MiR-148a表达变化对鼻咽癌细胞的增殖和凋亡影响。通过生物信息学软件分析预测MiR-148a的下游靶向调控基因表皮生长因子受体(EGFR),Real-time PCR和Western blot检测EGFR表达水平变化。结果:相对于正常鼻咽癌上皮组织细胞,MiR-148a在鼻咽癌细胞系CNE2中的表达水平显著降低。MTT实验、克隆形成实验和流式细胞术检测发现MiR-148a能够抑制细胞增殖,并促进细胞的凋亡。Real-time PCR和Western blot检测结果表明MiR-148a可降低EGFR的表达水平。结论:MiR-148a能够影响鼻咽癌细胞的增殖和凋亡,很有可能参与了鼻咽癌的发生发展过程。
[Abstract]:Aim: to investigate the expression of MiR-148a in nasopharyngeal carcinoma (NPC) cells and its effect on the biological function of nasopharyngeal carcinoma (NPC) cells. Methods: Real-time PCR was used to detect the expression of MiR-148a in nasopharyngeal carcinoma cell line CNE2. MTT assay, colony formation assay and flow cytometry were used to detect the effect of MiR-148a expression on the proliferation and apoptosis of nasopharyngeal carcinoma cells. The changes of EGFR expression level were predicted by bioinformatics software analysis, and EGFR expression level was detected by EGFR real-time PCR and Western blot, which was the downstream target regulatory gene of MiR-148a. Results: compared with normal nasopharyngeal carcinoma epithelial cells, the expression of MiR-148a in nasopharyngeal carcinoma cell line CNE2 was significantly decreased by MTT assay. Clone formation assay and flow cytometry showed that MiR-148a could inhibit the proliferation of nasopharyngeal carcinoma cell line CNE2. The results of cell apoptosis. Real-time PCR and Western blot analysis showed that MiR-148a could decrease the expression of EGFR. ConclusionMiR-148a can affect the proliferation and apoptosis of nasopharyngeal carcinoma cells and may be involved in the genesis and development of nasopharyngeal carcinoma.
