白藜芦醇诱导Hep-2人喉表皮样癌细胞自噬作用

【摘要】:目的:观察白藜芦醇对Hep-2细胞形态学的影响及诱导其自噬的可能机制。方法:40μM浓度白藜芦醇复合培养液作用于Hep-2细胞后,MDC染色和透射电镜分别检测自噬小体和自噬泡;Hoechest33258染色检测凋亡细胞;western blot分别分析自噬基因蛋白水平。结果:白藜芦醇增加LC3Ⅱ与LC3Ⅰ的比率,促进beclin1蛋白和mRNA的表达,细胞内的自噬泡和自噬小体数量也显著增加。用3MA和CQ抑制自噬则白藜芦醇对Hep-2细胞的促凋亡作用显著减弱。结论:白藜芦醇能够促进Hep-2细胞自噬;白藜芦醇诱导Hep-2细胞自噬的机制可能与促进LC3Ⅱ与LC3Ⅰ的表达、增强自噬基因beclin1蛋白的表达有关。
[Abstract]:Aim: to observe the effect of resveratrol on the morphology of Hep-2 cells and the possible mechanism of inducing autophagy.
