甘蔗叶多糖对鼻咽癌细胞株CNE2的体外抑制作用研究

【摘要】:目的:研究甘蔗叶多糖对鼻咽癌CNE_2细胞株的体外抑制作用,并初步探讨其作用机制。 方法:采用热水浸提及DEAE-纤维素层析柱法提纯甘蔗叶多糖,以MTT法观察其体外抗肿瘤活性,透射电子显微镜观察凋亡细胞的超微结构,采用流式细胞术检测肿瘤细胞凋亡率,实时荧光定量PCR法检测凋亡相关基因Bax、Bcl-2的变化。 结果:浓度为10ug/ml、20ug/ml、40ug/ml、80ug/ml、160ug/ml的甘蔗叶多糖作用后,CNE_2细胞抑制率及凋亡率均明显增加,且呈浓度依赖性。电镜下可见160ug/ml浓度的甘蔗叶多糖可使CNE_2细胞出现凋亡小体。荧光定量PCR技术测定Bax在CNE_2细胞中表达上调,并随着甘蔗叶多糖作用浓度的增加而升高;Bcl-2则在CNE_2细胞中表达下调,,且随着甘蔗叶多糖作用浓度的增加而降低。 结论:甘蔗叶多糖能明显抑制人鼻咽癌CNE_2细胞增殖,诱导细胞凋亡,可能通过提高Bax表达及降低Bcl-2的表达,从而发挥抗肿瘤作用。
[Abstract]:Aim: to study the inhibitory effect of sugarcane leaf polysaccharide on CNE2 cell line of nasopharyngeal carcinoma (NPC) in vitro and to explore its mechanism. Methods: the polysaccharides from sugarcane leaves were purified by hot water extraction and DEAE- cellulose chromatography. The in vitro antitumor activity was observed by MTT assay, the ultrastructure of apoptotic cells was observed by transmission electron microscope, and the apoptosis rate of tumor cells was detected by flow cytometry. The changes of apoptosis-related gene Baxon Bcl-2 were detected by real-time fluorescence quantitative PCR. Results: the inhibition rate and apoptosis rate of CNE2 cells were significantly increased after the treatment of sugar cane leaf polysaccharides at the concentration of 10ugrml / ml 20ugr / ml 40ugrml / ml ~ 80ugr / ml sugarcane leaf polysaccharide in a concentration-dependent manner. The inhibitory rate and apoptosis rate of CNE2 cells were increased in a concentration-dependent manner. Under electron microscope, 160ug/ml concentration of polysaccharides from sugarcane leaves could make CNE2 cells appear apoptotic bodies. The expression of Bax was up-regulated in CNE2 cells by fluorescence quantitative PCR, and the expression of Bcl-2 was down-regulated in CNE2 cells with the increase of polysaccharide concentration in sugarcane leaves, and decreased with the increase of polysaccharide concentration in sugarcane leaves. Conclusion: polysaccharide from sugarcane leaves can significantly inhibit the proliferation of CNE2 cells and induce apoptosis, which may play an anti-tumor role by increasing the expression of Bax and decreasing the expression of Bcl-2.
