基质金属蛋白酶-8对角膜基质胶原的作用研究
[Abstract]:Objective to investigate the effect of matrix metalloproteinase-8 (MMP-8) on cornea. Methods Fifteen healthy C57BL/6J mice were injected with 10 渭 L MMP-8 into the corneal stroma of the right eye as the experimental group, and the left eye was treated with the same amount of normal saline as the control group. Using two-photon microscope second harmonic imaging technique, the images were reconstructed by the software Imaris in vivo for 8 h. The image signal intensity was calculated for 8 h. The corneal turbidity was evaluated under slit lamp for 8 h. The levels of hydroxyproline in the cornea were measured. Results the signal intensities of corneal stromal fibers in the experimental group and the control group were 89.7 卤11.2U 85.3 卤7.0h and 14.5 卤3.4g 46.6 卤14.0h, respectively, which were 11.0 卤4.6 卤12.5h and 8h, respectively. The signal intensity of corneal stroma in the experimental group was significantly lower than that in the control group (P0.05) for 4h and 8h. The corneal signal intensity in the experimental group was significantly lower than that in the control group (P0.05) and the corneal signal intensity in the experimental group was significantly higher than that in the control group. The corneal hydroxyproline levels in the experimental group and the control group were (0.433 卤0.090), (卤0.590 卤0.133) 渭 g / mg, respectively, which were significantly lower than those in the control group (P 0.05). Conclusion MMP-8 can degrade and destroy the collagen of corneal stroma and decrease the transparency of cornea.
