咖啡酸苯乙酯抑制鼻咽癌的作用机制研究

  研究背景鼻咽癌(nasopharyngeal carcinoma,NPC)是一种特殊类型的头颈部恶性肿瘤,原发于解剖部位隐蔽的鼻咽部。NPC的发病有其独特的地理特点。NPC在东南亚,中国南方,尤其是广东、广西地区,年发病率约为30/10万,引发一系列严重的人群健康问题。而在美国或欧洲地区,其年发病率小于1/10万。NPC致癌过程是一个多步骤过程,目前发现NPC发病与EB病毒(Epstein-Barr virus,EBV)感染、遗传易感性、环境致癌因素密切相关。目前,NPC的治疗以放射治疗为主。随着调强放疗技术的发展,早期NPC患者通过放疗治疗5年生存率可以达到80%以上。然而,大部分NPC患者在初诊时已经处于中晚期,部分中晚期患者经过同步放化疗治疗后因肿瘤远处转移而治疗失败。因此,研发针对NPC的靶向药物,对放疗或化疗有增敏作用的药物,对提高中晚期NPC患者的疗效有积极意义。咖啡酸苯乙酯(caffeic acid phenethyl ester,CAPE)是一类多酚化合物。一些研究提示,CAPE具有抗癌,抗炎和免疫调节性质。其可以通过抑制肿瘤坏死因子(tumor necrosis fa...
