非酒精性脂肪肝大鼠肠道黏膜屏障功能的实验研究

【摘要】:目的肠道黏膜屏障功能紊乱与非酒精性脂肪肝(non-alcohol fatty liver disease,NAFLD)的发生关系密切,文中旨在通过动物模型的方法探讨NAFLD大鼠肠道黏膜屏障功能。方法 SD大鼠16只按随机数字表法分为正常组和NAFLD组,每组8只。正常组由普通饲料喂养,每100克普通饲料提供326 k Cal热量。NAFLD组大鼠由高脂饲料喂养,每100克高脂饲料提供453 k Cal热量建立NAFLD模型。采用HE染色检测肝病理,Elisa法和鲎试剂法分别检测肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1(Interleukin-1,IL-1)、内毒素,实时定量PCR法检测小肠闭锁小带蛋白ZO-1和连接蛋白Occluding的表达。结果 NAFLD组大鼠肝呈现典型的脂质沉积,血浆内毒素和血清TNF-α、IL-1较正常组显著增高,小肠ZO-1和Occluding表达较正常组显著降低(P0.05)。结论 NAFLD大鼠存在肠道黏膜屏障功能下降。
[Abstract]:Objective to investigate the relationship between intestinal mucosal barrier dysfunction and the occurrence of non-alcoholic fatty liver disease (NAFLD) in NAFLD rats.Methods Sixteen SD rats were randomly divided into normal group and NAFLD group with 8 rats in each group.The normal group was fed with normal diet, and the rats of NAFLD group were fed with high fat diet and every 100g high fat diet with 453k Cal calorie per 100g normal diet to establish NAFLD model.The expression of small intestinal atresia band protein (ZO-1) and connectin Occluding (Occluding) were detected by HE staining and Limulus lysate assay respectively. The expression of TNF- 伪, Interleukin-1 and endotoxin in small intestinal atresia band protein (ZO-1) and connectin Occluding (Occluding) were detected by real-time quantitative PCR.Results the liver of NAFLD group showed typical lipid deposition, plasma endotoxin and serum TNF- 伪 IL-1 were significantly higher than those in normal group, and the expression of ZO-1 and Occluding in small intestine was significantly lower than that in normal group.Conclusion the intestinal mucosal barrier function is decreased in NAFLD rats.
