枯草杆菌二联活菌对溃疡性结肠炎大鼠的作用及机制

【摘要】:目的探讨枯草杆菌二联活菌肠溶胶囊(美常安)对溃疡性结肠炎(UC)大鼠结肠促炎症性细胞因子白细胞介素(IL)-1β和细胞黏附分子CD44表达的影响及其作用机制。方法采用2,4,6-三硝基苯磺酸(TNBS)/乙醇法灌肠建立UC大鼠模型。将SD大鼠随机分成正常对照组、模型组、美常安治疗组。观察各组疾病活动指数(DAI)、结肠组织病理学评分(HPS)。造模及美常安干预7 d后处死大鼠,取结肠行HE染色,观察镜下结肠病理变化。RT-PCR法检测各组大鼠肠黏膜细胞因子IL-1βmRNA表达,流式细胞仪检测各组大鼠结肠组织中黏附分子CD44的含量。结果模型组较正常对照组大鼠结肠黏膜可见脓性分泌物、糜烂及溃疡,美常安干预后显著改善UC大鼠结肠黏膜的病理变化,DAI和HPS评分下降显著,并明显降低UC大鼠IL-1βmRNA和CD44的表达(P0.05)。结论枯草杆菌二联活菌可能通过下调UC大鼠IL-1βmRNA和CD44的表达而发挥干预作用。
[Abstract]:Objective to investigate the effect of Bacillus subtilis enteric-coated capsule (Meichang an) on the expression of interleukin-1 尾 (IL) 尾) and cell adhesion molecule (CD44) in colonic inflammatory cytokines (IL) 尾) and adhesion molecules (CD44) in rats with ulcerative colitis (UC). SD rats were randomly divided into normal control group, model group and mechangan treatment group. The disease activity index (DAI),) and colonic histopathology score (HPS).) were observed in each group. After 7 days of intervention, the rats were killed, the colons were stained with HE, and the expression of intestinal mucosal cytokine IL-1 尾 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). The content of adhesion molecule CD44 in colonic tissue of rats in each group was detected by flow cytometry. Results the mucosal purulent secretion, erosion and ulceration were observed in the model group compared with the normal control group. The pathological changes of colonic mucosa in UC rats were significantly improved after the intervention of Mechangan. The scores of Dai and HPS decreased significantly in the model group. The expression of IL-1 尾 mRNA and CD44 in UC rats was significantly decreased (P 0.05). Conclusion Bacillus subtilis combined with Bacillus subtilis may play an intervention role by down-regulating the expression of IL-1 尾 mRNA and CD44 in UC rats.
