荔枝核总黄酮对大鼠肝星状细胞MMP2及TIMP1表达的影响
[Abstract]:Aim: to observe the inhibitory effect of total flavone (TFL) on the proliferation of hepatic stellate cells (HSC) T 6) and its effect on the expression of MMP2,TIMP1. Methods: HSC-T6, cultured in vitro was randomly divided into control group and low, medium and high dose TFL groups. The control group was given 200渭 L medium containing 10s, and the low, middle and high dose groups were treated with 10s medium containing TFL160320640 渭 g/mL. The absorbance (A) of HSC-T6 at 490 nm was measured by MTT assay for 72 h. The expression of MMP2,TIMP1 mRNA and protein were detected by PCR and Western blotting assay at 72 h. Results: the proliferation of HSC-T6 was significantly increased in each group with the prolongation of the action time except for 48 minutes 72 hours in the high dose group of TFL (P 0.05). The proliferation of HSC-T6 was lower in 48 h group than that in control group (P 0.05), and the proliferation of HSC-T6 decreased gradually with the increase of TFL concentration at 72 h (all P 0.05) in the high dose group (P 0.05), and the proliferation of HSC-T6 in the high dose group was significantly lower than that in the control group at 48 h (P 0.05), but the proliferation of HSC-T6 in the high dose group was significantly lower than that in the control group (P 0.05). The relative expression of MMP2,TIMP1 mRNA and protein in the high dose group was lower than that in the control group at 72 h. The relative expression of MMP2,TIMP1 mRNA and protein decreased gradually with the increase of TFL concentration (all P 0. 05). ConclusionThree TFL can inhibit the proliferation of HSC-T6 in a time-and concentration-dependent manner, and its mechanism may be related to the down-regulation of MMP2 and TIMP1 expression.
