美洲大蠊水提取物对免疫性肝纤维化大鼠的保护作用

【摘要】:目的研究美洲大蠊水提取物对免疫性肝纤维化大鼠的保护作用及部分机制。方法将SD大鼠随机分为正常组、模型组、美洲大蠊30 mg/kg、60 mg/kg和120 mg/kg组和秋水仙碱物组,每组10只。免疫性肝纤维化模型通过大鼠腹腔注射猪血清制备,0.5 ml/kg,每周2次,共12周。造模期间给药组分别给予相应的受试药物,正常组和模型组分别给予等剂量的生理盐水。于造模给药结束后处死大鼠,测定肝脾指数;收集外周血,测定血清中丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、透明质酸(HA)、层粘蛋白(LN)、Ⅲ型前胶原酶(PCⅢ)和Ⅳ型胶原酶(CⅣ)含量;取肝脏,HE、Masson染色观察肝组织病理及胶原沉积变化;制备肝脏匀浆,ELISA法测定超氧化物歧化酶(SOD)活性和丙二醛(MDA)、谷光甘肽过氧化物酶(GSH-Px)含量;免疫组织化学和Western blot法测定肝组织中α-SMA、TGF-β1蛋白的表达。结果与模型组比较,美洲大蠊(30、60、120 mg/kg)能降低肝纤维化大鼠升高的肝脾指数,降低血清中ALT、AST、HA、LN、PCⅢ、CⅣ水平,降低肝组织MDA含量,提高SOD及GSH活性,减轻大鼠肝脏病理损伤程度,减少肝组织中α-SMA、TGF-β1的表达。结论美洲大蠊水提取物对猪血清诱导的免疫性肝纤维化大鼠有显著的保护作用,其机制可能与其抗氧化作用及抑制TGF-β1的表达有关。
