枸杞多糖防治非酒精性脂肪肝细胞水平的研究

【摘要】:目的:研究枸杞多糖(lycium barbarum polysaccharide,LBP)对肝细胞(LO2)脂肪变性的防治作用及其机制。方法:油酸诱导建立非酒精性脂肪变性肝细胞(non-alcoholic fatty liver disease,NAFLD)模型,分为正常对照组、脂肪变模型组、LBP预防组、LBP治疗组。48 h后光镜下观察细胞内脂质累积情况,并检测细胞内谷氨酸氨基转移酶(alanine aminostransferase,ALT)、门冬氨酸氨基转移酶(aspartate aminostransferase,AST)、甘油三酯(triglyceride,TG)含量,及脂联素与抵抗素的基因及蛋白表达情况。结果:光镜下可见模型组肝细胞内较正常对照组有大量橘红色脂滴(P=0.000),LBP预防组和治疗组与模型组比肝细胞内的脂滴明显减少(P=0.000、P=0.000)。LBP上调脂联素m RNA及蛋白表达(P=0.000、P=0.014),抑制抵抗素m RNA及蛋白的表达(P=0.000、P=0.000),降低肝细胞TG、ALT、AST(P=0.000、P=0.000、P=0.000)。结论:LBP可能通过上调脂联素m RNA及蛋白的表达,下调抵抗素m RNA及蛋白表达,从而有效防治NAFLD。
[Abstract]:Objective: to study the preventive and therapeutic effect of Lycium barbarum polysaccharide (lycium barbarum polysaccharide,LBP on steatosis of hepatocytes (LO2) and its mechanism. Methods: the model of nonalcoholic steatosis hepatocytes (non-alcoholic fatty liver disease,NAFLD) was induced by oleic acid and divided into normal control group, steatosis model group, LBP prevention group and LBP treatment group. 48 hours later, the intracellular lipid accumulation was observed under light microscope. The contents of intracellular glutamate aminotransferase (alanine aminostransferase,ALT), aspartate aminotransferase (aspartate aminostransferase,AST), glycerol (triglyceride,TG), and the expression of adiponectin and resistin gene and protein were detected.
