咪喹莫特治疗小鼠实验性结肠炎的效果研究

　　 背景:咪喹莫特是Toll样受体受体激动剂,可作为免疫佐剂,具有免疫调节作用,而溃疡性结肠炎与免疫紊乱有密切相关性。目的:本研究旨在探讨咪喹莫特(imiquimod,IMQ)对葡聚糖硫酸钠(DSS)诱导的小鼠急性结肠炎的疗效和可能机制。方法:将C57BL/6J小鼠随机分为健康空白对照组、DSS模型组、地塞米松(dexa-methasone,DXM)组(1.5mg/kg)及咪喹莫特组(IMQ,30mg/kg),除正常组饮食自由外,其余各组饮用3%DSS溶液连续7天构建实验性UC小鼠模型,同时每天分别给小鼠腹腔注射相应药物治疗,连续处理7天。治疗期间记录各组小鼠一般情况及死亡情况,在实验第八天牺牲小鼠,比较各组小鼠结肠长度、脾重,及DAI评分,HE染色观察小鼠结肠组织病理损伤变化,逆转录聚合酶链反应(RT-PCR)检测肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)mRNA表达水平;酶联免疫吸附法(ELISA)检测小鼠血清炎症因子 IL-6、TNF-α、IL-10 含量。结果:DSS成功诱发小鼠急性结肠炎,各实验组小鼠有明显腹泻,黏液脓血便,体重减轻等急性结肠炎表现,并且各实验组小鼠均有不同程度死亡。IMQ能有效改善DSS诱导的急性结肠炎症状,并能减轻小鼠结肠炎症严重程度。与DSS组相比,IMQ延缓了体重下降及结肠缩短(P0.01),增加了小鼠脾重(P0.05),下降疾病活动指数评分,降低结肠组织MPO活性(P0.01),并且炎症因子IL-6、TNF-α表达水平降低(P0.01),IL-10表达水平升高(P0.01)。结论:咪喹莫特对DSS诱发的小鼠实验性急性结肠炎有一定的治疗效果,主要通过诱导特异性免疫应答,增强机体免疫反应,降低炎症反应发挥作用。未来有望成为治疗溃疡性结肠炎的潜在药物。
