慢性乙型肝炎患者血清HBcAg的状态研究

目的研究慢性乙型肝炎（CHB）患者血清中HBcAg以什么方式存在。方法1、采用聚乙二醇（PEG）沉淀法沉淀血清样本中的HBV特异性循环免疫复合物并用胃蛋白酶解离复合物;2、通过免疫印迹检测解离物中的HBcAg复合物;3、用亲脂染料预染解离物,通过琼脂糖电泳检测脂质成分的存在。结果通过PEG沉淀,胃蛋白酶解离及Western Blot确定慢性乙型肝炎患者血清中有HBcAg特异性复合物存在,进一步预染样品后琼脂糖电泳证明HBcAg复合物上结合有脂质成分。结论证实了慢性乙型肝炎患者血清中有HBcAg特异性免疫复合物的存在,而且HBcAg是与脂质成分结合,说明血清中HBcAg是以脂复合物的形式存在着,从而推测慢性乙型肝炎患者血清中的HBcAg结合有脂膜成分,是膜表达HBcAg的残留,可能是免疫学系统攻击的靶抗原。
