RNA干扰抑制大鼠肝星状细胞CTGF表达的实验研究

目的构建针对结缔组织生长因子(connective tissue growth factor,CTGF)的RNA干扰重组质粒及其对照质粒,将其转染到体外培养的大鼠肝星状细胞(Hepatic Stellate Cells,HSC)中,进一步应用Real-time PCR方法检测CTGFmRNA的表达,观察RNA干扰对大鼠肝星状细胞CTGF表达的抑制作用,为临床基因治疗提供依据。方法①利用pGCsi载体构建介导CTGF短发夹RNA(shRNA)表达的质粒重组体,并通过基因测序分析进行鉴定。②将构建成功的4组重组质粒转染大鼠肝星状细胞株(HSC-T6),另设空白对照组,48 h后,共聚焦显微镜检测绿色荧光蛋白(green fluorescent protein,GFP)表达,分析转染效果。③检测CTGFmRNA表达水平,筛选出有效抑制CTGF表达的序列。④应用SPSS 17.0软件对数据进行方差分析。结果成功构建了能表达CTGFshRNA的三组重组体pGCsi-CTGFshRNA。pGCsi-CTGFshRNA成功转染到肝星状细胞中。与空白对照组相比,pGCsi-CTGFshRNA1组及pG...
