奥曲肽对肝硬化门脉高压大鼠肝脏血红素氧合酶-1表达的影响

  目的:肝硬化(hepatic cirrhosis)是各种慢性肝病发展的晚期阶段,是各种因素长期反复作用导致了肝细胞损伤,并引起肝星状细胞的活化,同时氧化应激以及多种细胞因子也参与了本病的发生发展过程。门静脉高压症(portal hypertension,PHT)是肝硬化失代偿期主要的临床表现之一,其引起的主要并发症食管胃底静脉曲张破裂出血严重威胁着患者的生命。血红素氧合酶(heme oxygenase, HO)是血红素代谢途径中的起始酶和限速酶,共有3种亚型,其中HO-1活性最高,广泛分布于全身组织,可被多种引起细胞氧化应激的损伤因素所诱导,HO-1及其催化产物胆绿素(biliverdin)、胆红素(biliubin)和CO(carbon monoxide,CO)在体内均有显著的抗炎、抗细胞增殖及调节细胞因子表达等重要作用。目前,奥曲肽(octreotide, OCT)作为临床上常用药物治疗食管胃底静脉曲张破裂出血,但奥曲肽在门脉高压阶段的作用机制尚不完全清楚。本研究观察奥曲肽降低门脉高压的作用是否调控了HO-CO系统,以探讨奥曲肽降门脉高压的可能机制,并进一步证实HO-CO系统在肝硬...
