丙型肝炎病毒NS2TP基因调节机制的研究

目的:NS2TP基因是抑制性消减杂交技术(SSH)筛选出的HCV NS2反式激活的基因之一。本实验中,我们对HCV NS2反式激活NS2TP基因从翻译水平和转录水平进行验证,进而定位NS2TP基因启动子的核心区域,并初步探究了NS2TP基因的转录调节机制。方法:(1) HCV NS2对NS2TP基因反式激活作用的验证。首先,运用pcDNA3.1(–)-NS2表达载体转染HepG2细胞,以pcDNA3.1(–)为阴性对照组。提取总蛋白、总RNA,然后分别从翻译水平,即Western Blot实验,和转录水平,即进行Real-time PCR实验。分析和研究NS2对NS2TP基因的反式激活作用。(2) NS2TP基因启动子的活性分析与核心区的确定。首先,成功构建pGL4.10-NS2TP启动子双荧光素酶报告基因表达载体系统,运用构建的不同截短型载体转染HepG2细胞,在对启动子活性进行分析的基础上,实验确定了NS2TP基因启动子活性的核心区域,同时也分析了NS2对NS2TP基因启动子的调节作用。(3) NS2TP基因转录调节因子CREB作用启动子核心区的验证。运用生物信息学技术分析了启动子...

Real Time PCR 扩增反应采用了如图 2-3 显示的 SYBR Green I 嵌合荧光法,即在反应体系中加入与双链 DNA 结合便可发出荧光的荧光染料 SYBR Green I,通过检测 PCR 反应液中的荧光信号强度,可以对目的基因进行准确定量,同时还可以测定扩增的目的 DNA 片段的融解温度。
