褪黑素对血小板衍生生长因子诱导的肝星状细胞增殖的影响及机制研究

目的:观察褪黑素对血小板衍生生长因子-BB(platelet-derived growth factor-BB,PDGF-BB)刺激的大鼠肝星状细胞(hepatic stellate cells,HSCs)增殖、PDGF-BB、血小板衍生生长因子受体-β(platelet-derived growth factor receptor-β,PDGFR-β)、α-平滑肌肌动蛋白(α-Smooth Muscle Actin,α-SMA)、细胞外信号调节激酶1/2(extracellular signal-regulated protein kinases1/2,ERK1/2)、磷酸化细胞外信号调节激酶1/2(phosphorylated extracellular signal-regulated kinase 1/2,p-ERK1/2)表达的影响,探讨褪黑素抗肝纤维化的分子机制。方法:以HSC-T6细胞为研究对象,分为对照组、模型组、褪黑素处理组(1 nmol/L、1μmol/L、0.1 mmol/L),培养24 h后,更换为无血清培养基,除对照组外均加入PDGF-BB(10 ng/mL)...
