副干酪乳杆菌N1115对乳鼠肠道发育影响研究

【摘要】:目的研究副干酪乳杆菌(Lactobacillus paracasei,Lp)N1115对小鼠生命早期肠道发育的影响。方法 C57BL/6小鼠出生后D1,开始喂食Lp N1115,检测体质量变化,至D14处死,用免疫组化法对绒毛宽度和隐窝深度,肠道上皮细胞增殖(Ki67核抗原)、分化(MUC2黏蛋白),紧密连接(ZO-1蛋白)进行测定。结果与对照组比较,Lp N1115干预组小鼠体质量在4 d后增加较快,体质量变化有显著性差异(P0.05)。免疫组化的结果显示,Lp N1115干预组小鼠的绒毛密度和隐窝深度增加,同时,K67阳性细胞和MUC2阳性细胞的数量较多,均有显著性(P0.05,P0.05)。ZO-1免疫组化结果显示,Lp N1115干预显著增加了ZO-1膜定位表达(P0.05)。结论早期应用Lp N1115可以促进肠上皮细胞增殖、分化及肠粘膜功能的建立,促进小鼠肠道发育。
[Abstract]:Objective to study the effect of Lactobacillus paracaseii LpN1115 on early intestinal development in mice. Methods C57BL/6 mice were fed with LpN1115 after birth and were killed by D14. The width of villi and the depth of crypt were determined by immunohistochemical method. The proliferation of intestinal epithelial cells was determined by Ki67 nuclear antigen, differentiation of MUC2 mucin and tight junction of ZO-1 protein. Results compared with the control group, the body weight of LpN1115 intervention group increased more rapidly after 4 days, and there was a significant difference in body mass change between LpN1115 group and control group (P 0.05). The results of immunohistochemistry showed that the density of villi and the depth of crypt were increased in LpN1115 group, and the number of K67 positive cells and MUC2 positive cells were more than that in LpN1115 group. The results of immunohistochemistry showed that LpN1115 significantly increased the expression of ZO-1 membrane. Conclusion early application of LP N1115 can promote the proliferation and differentiation of intestinal epithelial cells and the establishment of intestinal mucosal function, and promote the development of intestinal tract in mice.
