青春期双酚A暴露对小鼠认知功能和行为发育的影响

【摘要】:目的探讨青春期(即出生后28-56天,PND28-56)双酚A(Bispheno1A,BPA)暴露对ICR小鼠认知功能和行为的损害效应及其是否有持续影响。 方法选取4周龄ICR小鼠,随机均分为对照组、BPA(1、100mg/L)处理组。经玻璃瓶饮水染毒4周后,于成年早期采用Morris水迷宫试验来检测空间学习和记忆能力,采用旷场试验、黑白巷试验、高架十字迷宫试验来检测探索和焦虑相关行为,采用攻击行为实验来检测攻击行为。停止染毒,,饲养4周后,于成年期重复以上行为试验,来观察BPA暴露对小鼠认知功能、焦虑相关行为和攻击行为的损害效应是否具有持续影响。 结果 1.青春期和成年期阶段均无疾病、感染和死亡现象,体重的差异无统计学意义。 2.在Morris水迷宫实验中,BPA(100mg/L)处理组成年早期雌性小鼠的潜伏期和游泳距离显著延长(P 0.05),但雄鼠尚未观测到有差异;在成年期,各指标没有统计学差异。 3.在旷场实验中,与对照组相比较,BPA(1mg/L)处理组成年早期雌鼠的潜伏期有统计学差异(P 0.05);BPA(1mg/L)处理组成年早期雌鼠和成年期的雄鼠站立次数较对照组显著减少(P 0.05)。 4.在黑白巷实验中,各观测指标在成年早期差异没有统计学差异;BPA(100mg/L)处理组成年期雌鼠的黑巷时间与对照组相比差异有统计学意义(P 0.05)。 5.在攻击实验,BPA(100mg/L)处理组成年早期雄鼠的攻击潜伏期与对照组相比有差异(P 0.05);BPA(100mg/L)处理组成年早期和成年期雄鼠的接触时间与对照组相比均有差异(P 0.05)。 结论青春期BPA暴露对小鼠空间学习和记忆功能的损害呈现显著性别差异,对雌鼠损害效应更为显著;青春期BPA暴露对小鼠探索、焦虑和攻击行为带来持续性影响。
[Abstract]:Objective to investigate the effects of bisphenol A (BPA) exposure on cognitive function and behavior in ICR mice during puberty (28-56 days after birth). Methods four week old ICR mice were randomly divided into control group treated with BPA (1100mg / L). Four weeks after exposure to drinking water in glass bottles, Morris water maze test was used in early adulthood to test spatial learning and memory ability, open field test, black-and-white lane test, elevated cross maze test were used to detect behaviors related to exploration and anxiety. The attack behavior experiment was used to detect the attack behavior. After 4 weeks of feeding, the above behavioral tests were repeated in adult to observe whether BPA exposure had a lasting effect on cognitive function, anxiety related behavior and aggressive behavior in mice. Result 1. There was no disease, infection and death, and there was no significant difference in body weight between puberty and adulthood. In Morris water maze test, BPA (100 mg / L) significantly prolonged the latency and swimming distance of female mice in the early years of composition (P 0.05), but no significant difference was found in male mice, but there was no statistical difference among the indexes in adulthood. In the open field experiment, compared with the control group, there was a significant difference (P 0.05) in the incubation period of the female rats treated with BPA (1 mg / L) in the early stage of the constituent year (P 0.05), and the number of standing times of the male rats in the early stage of the composition year and the adult period was significantly decreased compared with the control group (P 0.05) (P 0.05). In the black-and-white lane experiment, there was no statistical difference between the observed indexes in the early adulthood. The black lane time of the female rats made up of BPA (100 mg / L) was significantly different from that of the control group (P 0.05). There was a significant difference in the incubation period between the male and the control group after BPA (100 mg / L) treatment (P 0.05). The exposure time of BPA (100 mg / L) was significantly different from that of the control group (P 0.05), and that of BPA (100 mg / L) was higher than that of the control group (P < 0.05). Conclusion the impairment of spatial learning and memory in mice exposed to pubertal BPA is significantly different from that of female mice, and the effects of puberty BPA exposure on exploration, anxiety and aggressive behavior in mice are persistent.
