2,4-二异氰酸甲苯酯暴露对小鼠血清中特异性IgG抗体水平的影响

【摘要】:[目的]探讨不同剂量和时间2,4-二异氰酸甲苯酯(2,4-TDI)气体暴露对小鼠血清中特异性Ig G抗体水平的影响。[方法]将清洁级(SPF)BALB/c雌性小鼠64只,按照染毒时间7 d和14 d随机分为两部分,共8组,每组8只,每部分分别设对照组及低(0.036 mg/m3)、中(0.14 mg/m3)、高(0.36 mg/m3)浓度暴露组,采用TDI连续动式吸入染毒,每天染毒4 h,记录体重,于末次染毒24 h采血分离血清,采用酶联免疫法检测特异性Ig G含量。[结果]暴露7 d时,3个暴露组小鼠血清特异性Ig G抗体含量(光密度值)分别为(0.14±0.008 4),(0.14±0.006 4)及(0.14±0.007 6),与对照组(0.14±0.009 3)相比,差异无统计学意义(P0.05);连续暴露14 d后小鼠血清中特异性Ig G抗体含量,各组间差异有统计学意义(F=11.41,P0.05),进一步两两比较分析,中、高暴露组血清中特异性Ig G抗体含量分别为(0.17±0.020 0)和(0.18±0.021 0),均高于对照组(0.14±0.004 6)(P0.05),高暴露组血清中特异性Ig G抗体含量高于低暴露组(0.15±0.012 0)(P0.05);中暴露组的暴露14 d小鼠血清中特异性Ig G抗体含量为(0.17±0.020 0),高于暴露7 d(0.14±0.006 4)(P0.05);高暴露组的暴露14 d小鼠血清中特异性Ig G抗体含量为(0.18±0.021 0),高于7 d(0.14±0.007 6)(P0.05)。[结论]TDI气体连续暴露浓度及时间均影响小鼠血清中特异性的Ig G抗体水平的表达。
[Abstract]:[objective] to investigate the effect of 2o 4 diisocyanate gas exposure at different doses and time on the level of specific IgG antibody in serum of mice. [methods] Sixty-four clean grade (SPF) BALB/c female mice were randomly divided into 8 groups according to the exposure time of 7 days and 14 days. Each part was divided into control group and low (0.036 mg/m3), middle (0.14 mg/m3), high (0.36 mg/m3) exposure group. The serum was continuously inhaled by TDI for 4 hours daily, and the body weight was recorded. The serum samples were collected for 24 hours after the last exposure. The specific IgG content was detected by enzyme linked immunosorbent assay (Elisa). [results] after 7 days of exposure, the specific IgG antibody levels (optical density) in serum of the three exposure groups were (0.14 卤0.008 4), (, 0.14 卤0.006 4) and (0.14 卤0.007 6), respectively, compared with those of the control group (0.14 卤0.009 3). There was no significant difference (P0.05); after 14 days of continuous exposure, the content of specific IgG antibody in serum of mice was statistically significant (FG 11.41 P 0.05). The specific IgG antibody levels in the serum of the high exposure group were (0. 17 卤0.020) and (0. 18 卤0.021), respectively, which were higher than those of the control group (0. 14 卤0.004. 6) (P0.05), the level of serum specific IgG antibody in the high exposure group was higher than that in the low exposure group (0. 15 卤0.012) (P0.05), and that in the middle exposure group was 14 days after exposure (P0.05). The specific IgG antibody content was (0.17 卤0.020), higher than that in the 7th day (0.14 卤0.006 4) (P0.05), and the specific IgG antibody level in the serum of the mice exposed to high exposure for 14 days was (0.18 卤0.021), higher than that in the 7th day (0.14 卤0.007 6) (P0.05). [conclusion] the concentration and time of continuous exposure of TDI affected the expression of specific IgG antibody in serum of mice.
