八珍加肉桂补骨脂汤对运动疲劳大鼠海马细胞凋亡和bcl-2、bax蛋白表达的影响

【摘要】:[研究目的]:通过对大鼠运动疲劳模型的建立,从大鼠海马细胞凋亡情况以及bcl-2、bax蛋白表达水平变化的角度来研究八珍加肉桂补骨脂汤对运动疲劳的保护作用和具体机制。[研究方法]:采用SD雄性大鼠45只,体重(185±10)克,根据实验要求将大鼠随机分为3组:对照组(S,N=15)、模型组(E,N=15)、中药加运动组(CE,N=15)。除去对照组不做任何训练之外,其它2组按计划进行7周逐级增加负荷的跑台运动训练,以此建立慢性运动性疲劳模型。对照组无任何的运动训练,正常饲养;模型组在每天训练结束后半小时之内,灌胃一次安慰剂生理盐水;中药加运动组训练结束后半小时之内,灌胃一次复方中药八珍加肉桂补骨脂汤。[研究结果]:(1)大鼠海马组织5-羟色胺、多巴胺含量的比较:和S组比较,E组的大鼠海马组织5-HT的含量明显上升,而多巴胺的含量则明显下降(P0.05);和E组比较,CE组的大鼠海马组织5-HT的含量明显下降,而多巴胺的含量明显上升(P0.05)。(2)大鼠海马组织细胞凋亡情况的比较:和S组比较,E组大鼠海马组织中凋亡的细胞数量大大增加(P0.05),具有显著性差异;和E组比较,CE组大鼠海马组织中凋亡细胞的数量大大减少(P0.05),具有显著性差异。(3)大鼠海马细胞bcl-2蛋白表达的比较:和S组比较,E组大鼠海马细胞内bcl-2蛋白表达水平较明显上升(P0.05);和E组比较,CE组大鼠海马细胞内bcl-2蛋白表达水平明显上升(P0.05)。(4)大鼠海马细胞bax蛋白表达的比较:和S组比较,E组大鼠海马细胞内bax蛋白表达水平明显上升(P0.05);和E组比较,CE组大鼠海马细胞内bax蛋白表达水平明显下降(P0.05)。[结论]:(1)八珍加肉桂补骨脂汤通过对多巴胺、5-羟色胺等中枢神经递质指标的调节,达到延缓了运动性中枢疲劳的发生。(2)八珍加肉桂补骨脂汤在一定程度上抑制了运动疲劳大鼠海马组织内的细胞凋亡,对中枢神经细胞起到了保护作用。(3)八珍加肉桂补骨脂汤通过对bcl-2和bax蛋白表达水平的调节,抑制了细胞凋亡,对中枢神经细胞起到了保护作用,从而在一定程度上缓解了运动性中枢疲劳的发生。
[Abstract]:[objective]: to establish a model of exercise fatigue in rats. From the point of view of apoptosis of rat hippocampal cells and the change of bcl-2nbax protein expression, the protective effect and specific mechanism of Bazhen plus cinnamon psoralen decoction on exercise fatigue were studied. [methods]: 45 SD male rats, weight of 185 卤10 g, were used. The rats were randomly divided into three groups according to the requirements of the experiment: control group, model group, Chinese herbal medicine plus exercise group. Except for the control group without any training, the other two groups were trained for 7 weeks in order to increase the load step by step. The model of chronic exercise fatigue was established. The control group had no exercise training and was raised normally. The model group was given a placebo saline once a day within half an hour after the training, and the Chinese medicine plus exercise group was fed within half an hour after the training. The contents of 5-hydroxytryptamine and dopamine in hippocampal tissue of rats were compared: compared with group S, the content of 5-HT in hippocampal tissue of rats in group E was obviously increased, and the content of 5-HT in hippocampus of rats in group E was significantly higher than that in group S, and the content of 5-HT in hippocampus of rats in group E was significantly higher than that in group S. Compared with group E, the content of 5-HT in hippocampal tissue of rats in CE group was significantly decreased, and the content of 5-HT in hippocampus of rats in group E was significantly lower than that in group E, while the content of dopamine decreased significantly. Compared with group S, the number of apoptotic cells in hippocampal tissue of rats in group E was significantly increased, and there was significant difference in the number of apoptotic cells between group E and group S; Compared with group E, the number of apoptotic cells in hippocampal tissue of rats in CE group decreased significantly (P 0.05) and the expression of bcl-2 protein in hippocampal cells of rats was significantly different. Compared with group S, the expression level of bcl-2 protein in hippocampal cells of rats in group E was higher than that in group S. Compared with group E, the expression of bcl-2 protein in hippocampal cells of rats in CE group was significantly higher than that in group E, and the expression of bax protein in hippocampal cells of rats in group E was significantly higher than that in group S. The expression level of bax protein in hippocampal cells of rats in group E was significantly higher than that in group S. Compared with group E, the expression level of bax protein in hippocampal cells of rats in the CE group decreased significantly. [conclusion] Bazhen and Pengguzhi decoction can regulate the central neurotransmitters, such as dopamine 5-hydroxytryptamine, by regulating the expression of P0.05 protein in the hippocampus of the rats in the CE group. [conclusion] Bazhen + Psoralea decoction can regulate the neurotransmitters, such as dopamine serotonin, and so on. Bazhen plus cinnamon psoralen decoction inhibited apoptosis in hippocampus of exercise fatigue rats to some extent. Bazhen plus cinnamon psoralen decoction inhibits apoptosis and protects central nervous cells by regulating the expression of bcl-2 and bax proteins. To a certain extent, it alleviates the occurrence of exercise-induced central fatigue.
