碳离子束和X射线对头颈癌患者与健康人离体外周血淋巴细胞亚群影响的对比研究

【摘要】:目的:对比研究碳离子束和X射线对健康人与头颈部恶性肿瘤患者离体外周血淋巴细胞亚群影响,了解不同射线、不同放射治疗剂量对健康人和恶性肿瘤患者离体外周血淋巴细胞亚群分布的差异。方法:在采血后8小时内分别行6MV-X线1Gy、2Gy照射及~(12+)C离子束1Gy E、2Gy E照射;应用美国BD公司的四色荧光标记抗人CD分子单克隆抗体和FACSVerse流式细胞仪,检测外周血淋巴细胞亚群。结果:各T淋巴细胞亚群CD3~+、CD4~+、CD8~+、CD4~+/CD8~+及NK细胞、B细胞无显著差异(P0.05),但CD4~+/CD8~+在健康人离体血中下降更为明显。结论:对健康人和头颈部鳞癌患者的离体外周血行6 MV-X线1 Gy、2 Gy照射及~(12+)C离子束1Gy E、2Gy E照射,两种射线对T淋巴细胞亚群的影响无明显差异,健康人和头颈部鳞癌患者间也无明显差异。结果对临床中放射治疗放射方法的选择具有重要参考价值。
[Abstract]:Objective: to compare the effects of carbon ion beam and X ray on lymphocyte subsets in peripheral blood of healthy people and patients with malignant tumor of head and neck. The distribution of lymphocyte subsets in peripheral blood of healthy persons and malignant tumor patients was different with different doses of radiotherapy. Methods: within 8 hours after blood collection, 6MV-X ray irradiation of 1 Gy ~ 2Gy and ~ (12) C ~ + beam of 1 Gy E ~ (2 Gy) were carried out respectively. The monoclonal antibody against human CD molecule and FACSVerse flow cytometry were labeled with four colors fluorescence from BD Company of USA. Results: there was no significant difference between CD3 ~ + CD4 ~ + CD8 ~ + CD4 ~ / CD8 ~ + T lymphocyte subsets and NK cell B cells, but CD4 ~ / / CD8 ~ + decreased more obviously in the blood of healthy people in vitro. Conclusion: in healthy people and head and neck, there is no significant difference between them. In vitro peripheral blood of patients with squamous cell carcinoma was irradiated with 1 Gy / 2 Gy of 6 MV-X line and 1 Gy / 2 Gy of E ~ (2 Gy) with ~ (12) C ion beam. There was no significant difference between the two kinds of radiation on T lymphocyte subsets, and there was no significant difference between healthy persons and patients with head and neck squamous cell carcinoma. The results have important reference value for the choice of radiotherapy methods in clinic.
