富氢水抑制离心运动后骨骼肌线粒体氧化应激和炎症反应

【摘要】:背景:富氢水能否用于离心运动肌损伤的防护及其相关机制尚不清楚。目的:观察富氢水对离心运动大鼠骨骼肌线粒体氧化应激及炎症反应的影响,并探讨富氢水作用的相关信号通路。方法:将40只SD大鼠随机分为对照组、离心运动组、离心运动+生理盐水组和离心运动+富氢水组。后3组大鼠连续5d进行下坡跑,跑台速度为16-18m/min,坡度-16°,90min/d,共5d。离心运动+富氢水组大鼠在每次运动后即刻腹腔注射饱和富氢水(10mL/kg);离心运动+生理盐水组大鼠注射生理盐水。结果与结论:离心运动同时给予富氢水显著上调Sirtuin-3(Sirt3)表达,提高线粒体膜电位和锰超氧化物歧化酶活性,抑制线粒体活性氧生成和线粒体DNA氧化损伤,降低炎性因子NOD样受体热蛋白结构域相关蛋白3和白细胞介素1β表达。表明富氢水除可直接清除活性氧外,还可通过上调SIRT3表达,提高线粒体能量代谢和抗氧化能力,从而抑制离心运动诱导的线粒体氧化应激和继发的炎症反应,实现骨骼肌保护效应。
[Abstract]:Background: it is unclear whether hydrogen-rich water can be used in the protection of eccentric muscle injury and its related mechanism. Aim: to investigate the effects of hydrogen rich water on oxidative stress and inflammatory response of skeletal muscle mitochondria during centrifugation in rats, and to explore the signal pathways related to the action of hydrogen rich water. Methods: forty Sprague-Dawley rats were randomly divided into control group, centrifuge exercise group, centrifuge normal saline group and hydrogen rich water group. The latter three groups were followed by downhill running for 5 days. The running speed was 16-18 m / min, and the slope was -16 掳/ 90 min / d for 5 days. Rats in the group of hydrogen-rich water in centrifugation group were injected with saturated hydrogen rich water 10 mL / kg immediately after each exercise, and rats in the group of normal saline in centrifugal exercise group were injected with normal saline. Results & Conclusion: at the same time, the hydrogen-rich water increased the expression of Sirtuin-3n Sirt3, increased the mitochondrial membrane potential and the activity of manganese superoxide dismutase, inhibited the production of reactive oxygen species in mitochondria and oxidative damage of mitochondrial DNA. The expression of inflammatory factor NOD like receptor domain associated protein 3 and interleukin 1 尾 was decreased. In addition to scavenging reactive oxygen species directly, hydrogen-rich water can also increase the energy metabolism and antioxidant capacity of mitochondria by upregulating the expression of SIRT3, thus inhibiting the oxidative stress and the secondary inflammatory response induced by centrifugal exercise. The protective effect of skeletal muscle was realized.
