4周间歇性禁食对大鼠骨骼肌质量及自噬的影响

【摘要】:目的:探讨4周间歇性禁食后大鼠体重、体脂及骨骼肌质量的变化与大鼠骨骼肌自噬的关系,为间歇性禁食提供理论依据。方法:20只雄性SD大鼠随机分为对照组(Con组)和间歇性禁食组(IF组),每组各10只。采取每周三、周五间歇性禁食方案,每周按时记录大鼠体重,4周干预后双能X线吸收测量法(DEXA)分析体脂含量,之后分离双侧比目鱼肌称量湿重,免疫荧光法检测比目鱼肌laminin蛋白体现肌肉横截面积,透射电镜观察比目鱼肌自噬泡形态,Western Blot检测自噬相关蛋白LC3、p62及调控蛋白AMPKα、p-AMPKα、ULK1的表达情况。结果:4周间歇性禁食后,间歇性禁食组大鼠体重与体脂含量显著低于对照组(P0.01),但各组比目鱼肌湿重及肌纤维横截面积并无明显差异(P0.05)。间歇性禁食组比目鱼肌AMPKα、p-AMPKα、ULK1蛋白表达水平明显高于对照组(P0.01),间歇性禁食组比目鱼肌内自噬标记LC3-Ⅱ/LC3-Ⅰ水平明显高于对照组,p62表达水平明显低于对照组,均具有非常显著性差异(P0.01)。结论:4周间歇性禁食明显降低体脂水平,具有控制体重的作用,可激活AMPK-ULK1通路适度促进骨骼肌自噬,以维持骨骼肌质量,可作为一种潜在的"减肥"方法进行深入研究。
[Abstract]:Objective: to investigate the relationship between the changes of body weight, body fat and skeletal muscle mass and the autophagy of skeletal muscle in rats after intermittent fasting for 4 weeks, and to provide a theoretical basis for intermittent fasting. Methods Twenty male Sprague-Dawley rats were randomly divided into control group (Con group) and intermittent fasting group (10 rats in each group). The rats' body fat content was measured by dual energy X-ray absorptiometry after 4 weeks of intervention, and the wet weight of bilateral soleus muscle was separated. The cross sectional area of soleus muscle laminin protein was detected by immunofluorescence, and the expression of autophagy associated protein LC3p62 and the regulatory protein AMPK 伪 p-AMPK 伪 mULK1 in soleus muscle were observed by transmission electron microscope (TEM). Results after 4 weeks of intermittent fasting, the body weight and body fat content of rats in the intermittent fasting group were significantly lower than those in the control group (P 0.01), but there was no significant difference in wet weight of soleus muscle and cross sectional area of muscle fiber in each group.
