运动、EGCG和左旋肉碱对肥胖大鼠脂肪代谢和能量代谢的影响

【摘要】:研究目的:观察运动以及运动联合EGCG或(和)左旋肉碱的减肥效果,探索其对脂肪代谢和能量代谢的影响。 研究方法:以4周龄雄性SD大鼠为研究对象,高脂饲料诱导饮食型肥胖模型。然后进行6周减肥干预实验:将肥胖大鼠随机分为肥胖模型组(F,n=6)、单纯运动组(S,n=6)、运动联合EGCG组(E,n=6)、运动联合LC组(L,n=6)、运动联合EGCG和LC(EL,,n=6),仍然给予高脂饲料;运动采取中低强度跑台运动方式,EGCG和LC按80mg/kg bw剂量灌胃。实验结束后测试各组血脂、肝脏脂酶活性以及附睾脂肪HSL和褐色脂肪UCPl蛋白表达水平。 研究结果:1)与F组相比,各干预组大鼠体重明显降低(P0.05):其中EL组体重较S组明显下降(P0.05),其余各组之间没有差异(P0.05)。2)各干预组血清TG显著低于F组(P0.01),TC呈降低趋势(P0.05), HDL-C为增加趋势(P0.05)。与S组相比,E组、L组、EL组TG显著降低(P0.01),而EL组TC和HDL-C明显改善(分别为P0.05,P0.01),其余各组之间没有明显区别(P0.05)。3)与F组相比,E组、L组、EL组显著提高了肝脏脂酶活性(P0.01,P0.05,P0.01):其中E组、EL组的脂酶活性明显高于S组(P0.05),同时与L组相比亦明显增加(P0.05)。4)各干预组HSL蛋白水平显著高于F组(P0.01)。E组HSL蛋白表达高于S组(P0.05),其他各组之间没有明显差异(P0.05)。5)与F组相比,S组和L组UCPl蛋白表达没有明显差异(P0.05),仅E组和EL组明显提高了褐色脂肪UCP1蛋白表达(P0.05)。 研究结论:1)6周运动或运动联合EGCG或(和)LC均可降低肥胖大鼠的体重,提高肝脏脂酶活性并改善血脂代谢,增强脂肪组织HSL蛋白表达促进脂肪动员和氧化利用。2) EGCG表现出提高褐色脂肪UCP1蛋白表达的能力,有氧运动或LC对BAT生热作用没有影响。3) EGCG在提高肝脂酶、改善血脂和增强HSL蛋白表达方面与运动有协同促进作用,而LC仅协同促进运动对血脂的改善。4)在减轻体重方面运动联合EGCG和LC组有效叠加了运动、LC的独立效果。
[Abstract]:Objective: To observe the effect of exercise and exercise combined with EGCG or / or L-carnitine on weight loss and to explore its effects on fat metabolism and energy metabolism.
Study methods: 4 weeks old male SD rats were used as the research object, high fat diet induced diet obesity model. Then 6 weeks of weight loss intervention experiment were carried out: obese rats were randomly divided into obesity model group (F, n=6), simple exercise group (S, n=6), exercise combined EGCG group (E, n=6), exercise combined LC group (L, n=6), exercise joint EGCG and concerned, still given High fat diet; exercise with medium and low intensity treadmill exercise, EGCG and LC gavage at 80mg/kg BW dose. After the experiment, blood lipid, liver lipase activity, and epididymal fat HSL and brown fat UCPl protein expression level were tested.
The results were as follows: 1) compared with the F group, the weight of the rats in the intervention group was significantly lower (P0.05): the weight of the EL group was significantly lower than that in the S group (P0.05), and there was no difference between the other groups (P0.05).2) and the serum TG was significantly lower than the F group (P0.01) in the intervention groups (P0.01), and the TC showed a decreasing trend (P0.05). In group EL, TC and HDL-C were significantly improved (P0.05, P0.01), and there was no significant difference between the other groups (P0.05).3). Compared with the F group, the activity of livers was significantly increased in the E group and L group, and the livers activity of the group was significantly higher than that in the group of E. The level of pre group HSL protein was significantly higher than that in group F (P0.01).E group, HSL protein expression was higher than that of S group (P0.05). There was no significant difference between the other groups (P0.05).5). Compared with F group, there was no significant difference between S group and L group.
Conclusions: 1) 1) 6 weeks of exercise or exercise combined with EGCG or (and) LC can reduce the weight of obese rats, improve liver lipase activity, improve blood lipid metabolism, enhance the expression of HSL protein in adipose tissue and promote lipid mobilization and oxidative utilization of.2) EGCG shows the ability to improve the expression of brown fat UCP1 protein, aerobic exercise or LC to BAT No effect on.3) EGCG has a synergistic effect on improving liver lipase, improving blood lipid and enhancing the expression of HSL protein, while LC only synergistically promotes exercise to improve blood lipid.4) the exercise combined with EGCG and LC groups effectively combined exercise and LC's independent effect.
