有氧运动对D-半乳糖诱导的脑衰老大鼠前额叶SYP、PSD-95表达的影响
[Abstract]:Aim: to observe the effect of aerobic exercise on the expression of presynaptic protein 95 (PSD-95) in prefrontal synaptophysin (SYP), in brain of aging rats induced by D-galactose (D-gal). To explore the potential mechanism of learning and memory impairment during aerobic exercise delaying brain aging. Methods: Thirty-six 3-month-old SD rats were randomly divided into normal saline control group (group C), D-gal group (group D) and D-gal plus aerobic exercise group (DE group) with 12 rats in each group. Rats in the DE group were treated with intraperitoneal injection of D-gal (100 mg/kg/d) daily for 6 weeks, while rats in the DE group underwent swimming training during D-gal injection (1 h / d / 6 d / w). Then the spatial learning and memory ability of rats was measured by Morris water maze, and the expression of SYP,PSD-95 protein and m RNA in the prefrontal lobe of rat brain were detected by immunofluorescence, Western Blot and Real Time-PCR techniques, respectively. Results: (1) the escape latency of the rats in the Morris water maze experiment decreased gradually during the navigation training. Group C and group DE showed the best results on the 3rd day of training, but there was no significant change after that. The average escape latency of group D was significantly longer than that of group C and DE (P0.05), but there was no significant difference between the two groups (P0.05). In the space exploration experiment, the times of crossing the original platform area in group D within 60 seconds were significantly lower than those in group C and DE (P0.05), and the latter two groups had a longer stay in the quadrant of the original platform (P0.05). (2) compared with group C. The expression of SYP protein and SYP m RNA in prefrontal lobe of D group was significantly lower than that in D group (P0.01), DE group was significantly higher than that in D group (P0.05). The expression of PSD-95 protein and its m RNA in prefrontal lobe of group D were significantly lower than those in group C (P0.01P 0.05). There was no significant difference in PSD-95 m RNA expression between DE group and D group, but the expression of PSD-95 protein in DE group was significantly higher than that in D group (P0.05). Conclusion: aerobic exercise can inhibit the expression of synaptophysin SYP,PSD-95 in the prefrontal lobe of rats during aging induced by D-gal to some extent, and reduce the degree of decrease in the expression of synaptophysin in the prefrontal lobe of rats. This may be one of the molecular mechanisms by which aerobic exercise delays the decline of brain aging learning and memory.
