人白细胞中SIRT1和FoxO3a mRNA表达的随龄变化研究

【摘要】:研究背景在目前的法医学实践中,个体年龄的推断主要是根据骨骼、牙齿等骨性指标的随龄变化特征,应用法医人类学模型计算来完成的。该技术已经比较成熟,且误差也相对较小。但在遇到骨骼、牙齿等检材缺失的案例时,其应用就受到了极大的限制。随着分子生物学的发展,通过检测端粒长度、线粒体DNA损伤程度及DNA甲基化水平等推断个体年龄已成为研究热点,但因种种原因,目前尚难应用于法医学实践。近年来研究发现,沉默信息调节蛋白1(SIRT1)和叉头蛋白O亚家族3a(FoxO3a)在细胞衰老和人体长寿中发挥重要作用,这也提示SIRT1和FoxO3a的表达可能与年龄存在密切关系。因此如果能建立SIRT1和FoxO3a基因表达与年龄之间的数学模型,将有可能实现通过这两种基因表达的丰度测定来推断个体年龄,有望为法医学中的年龄推断提供新的技术手段。 目的通过检测比较不同年龄段个体SIRT1和FoxO3a mRNA的表达水平,并分析其随龄变化趋势,为进一步建立SIRT1和FoxO3a mRNA表达与年龄之间的数学模型奠定基础。 方法采集80个健康无关个体血样,按年龄分为8组:≤9岁、10~19岁、20~29岁、30~39岁、40~49岁、50~59岁、60~69岁及≥70岁组,每组10人。提取总RNA后,利用实时定量逆转录聚合酶链反应(RT-qPCR)技术,检测人白细胞中SIRT1和FoxO3a mRNA的表达情况,,用SPSS软件对数据资料进行统计分析。 结果⑴人白细胞SIRT1mRNA相对表达水平在≤9岁、10~19岁、20~29岁、30~39岁、40~49岁和50~59岁组之间均无显著性差异(P>0.05),而60~69岁和≥70岁组分别与上述各组有显著性差异(P<0.01),且此两组之间也有显著性差异(P<0.01)。白细胞SIRT1mRNA相对表达水平与年龄正相关(r=0.708,P<0.01),以年龄分组为自变量,SIRT1mRNA相对表达水平为因变量,进行曲线拟合得Y=0.005X3-0.002X2-0.004(R2=0.884,P<0.01);⑵人白细胞FoxO3a mRNA在≤9岁与10~19岁组之间、≤9岁与20~29岁组之间,10~19岁与20~29岁组之间,20~29岁与30~39岁组之间,30~39岁与40~49岁组之间相对表达水平无显著性差异(P>0.05),其余任何两组之间白细胞FoxO3a mRNA相对表达水平有显著性差异(P<0.05)。白细胞FoxO3amRNA相对表达水平与年龄正相关(r=0.841, P<0.01),以年龄分组为自变量,FoxO3a mRNA相对表达水平为因变量,进行曲线拟合得Y=0.001e0.418x(R2=0.935,P<0.01)。 结论⑴人白细胞SIRT1mRNA在健康个体年龄㩳60岁时表达量非常低,而在≥60岁时高表达;⑵不同年龄组健康个体白细胞FoxO3a mRNA表达随年龄的增长而升高;⑶人白细胞FoxO3a mRNA表达水平有望成为年龄推断的有用指标之一。
[Abstract]:Background in the current forensic practice, the estimation of individual age is mainly based on the characteristics of bone, teeth and other bone indicators with age, using forensic anthropology model to complete the calculation. The technology has been more mature, and the error is relatively small. However, in the case of missing bone, teeth and other materials, its application is greatly limited. With the development of molecular biology, the estimation of individual age by detecting telomere length, mitochondrial DNA damage and DNA methylation has become a hot topic. However, it is difficult to apply it to forensic practice for various reasons. In recent years, silencing information regulatory protein 1 (SIRT1) and fork head protein O subfamily 3a (FoxO3a) have been found to play an important role in cell senescence and longevity, which suggests that the expression of SIRT1 and FoxO3a may be closely related to age. Therefore, if a mathematical model between SIRT1 and FoxO3a gene expression and age can be established, it will be possible to infer individual age by determining the abundance of these two genes, which may provide a new technique for age estimation in forensic medicine. Objective to compare the expression levels of SIRT1 and FoxO3a mRNA in individuals of different ages and analyze their trends with age so as to establish a mathematical model between SIRT1 and FoxO3a mRNA expression and age.
