CsA干预下长时间耐力运动对大鼠Caveolin-3及相关指标的影响

【摘要】:研究目的:在CsA干预条件下,长时间大强度的耐力运动对细胞膜caveolin-3含量的影响情况。结合一些生化指标和细胞内离子浓度的变化,探讨caveolin-3在耐力运动中参与对运动引起细胞蛋白代谢、对细胞信号通路的影响及caveolin-3自身变化机制,为耐力运动造成的细胞结构的变化以及所引起的疲劳快速恢复提供理论依据。 研究方法:研究对象选用6周龄大的SD雄性大鼠40只,随机分成安静组(C)、耐力训练组(T)、环孢霉素组(S)和环孢霉素训练组(ST),每组10只。适应性训练两周后,以28m/s,10°坡度,每天训练1小时,每周休息1天的强度训练13周,11-13周ST组和S组注射CsA。腹主动脉取血,取腓肠肌、比目鱼肌、趾长伸肌。免疫组化法测Caveolin-3含量;荧光指示法测定Ca~(2+);采用速率法测定血液中CK、LDH及组织LDH的含量;硝酸还原酶法测定组织中NO、NOS的含量。 实验结果: (1)长时间大强度运动后,环孢霉素组(S)大鼠细胞内Ca~(2+)浓度与安静组(C)相比无显著性差异(p0.05);耐力训练组(T)极其显著高于安静组(C)(p0.01);环孢霉素耐力训练组(ST)显著高于安静组(C)(p0.05)。 (2)NO测试结果为:环孢霉素组(S)显著高于安静组(C)(p0.05);耐力训练组(T)与安静对照组(C)相比,无显著性差异(p0.05);环孢霉素耐力训练组(ST)极其显著高于安静组(C)(p0.01),显著高于运动对照组(p0.05)。 (3)总NOS测得结果为:环孢霉素组(S)与安静组(C)相比,TNOS含量极其显著增高(p0.01);耐力训练组(T)与安静组(C)相比,TONS含量显著升高(p0.05);环孢耐力训练组(ST)与安静组(C)相比,无显著性差异(p0.05)。 (4)CK测试结果:各组间无显著性差异(p0.05)。 (5)血液中LDH测试结果为环孢霉素组显著高于安静组(p0.05);其它组间相比无显著性差异(p0.05)。组织中测试结果为:各组间无显著性差异(p0.05)。 (6)Caveolin-3的测试结果显示:趾长伸肌中的含量由高到低的顺序为C组、ST组、T组、S组;比目鱼肌中的含量由高到低顺序为C组、T组、S组、ST组。 实验结论: 1.环孢霉素A会造成骨骼肌细胞膜的损伤。 2.长时间耐力运动后细胞内Ca~(2+)浓度上升,单独CsA干预对胞浆Ca~(2+)浓度变化影响不大。耐力运动与CsA共同作用下,,细胞内Ca~(2+)浓度上升。耐力运动干预条件对胞浆内Ca~(2+)浓度的影响明显高于CsA干预条件。 3.CsA干预条件下,骨骼肌组织中NOS与NO变化一致,都会升高。耐力训练下,肌组织中NOS会升高,NO的生成受到影响。CsA与耐力运动共同影响下,NOS变化不明显,NO含量增加。 4.CsA干预下,组织中LDH含量上升,CK变化恢复到安静时水平。与CK相比,LDH恢复的更慢,反映损伤的时间更长。 5.长时间耐力运动、CsA干预与二者协同作用都能够造成不同纤维类型肌肉中Caveolin-3含量的降低,影响Caveolin-3介导的信号通路。CsA对比目鱼肌为代表的慢肌纤维的影响较以趾长伸肌为代表的快肌纤维的影响更为显著。
[Abstract]:Objective: To study the effect of long-time and long-time endurance exercise on the content of caveolin-3 in cell membrane under the condition of CsA intervention. In combination with that change of the biochemical index and the ion concentration in the cell, the influence of cave1-3 on the cellular protein metabolism, the effect on the cell signal path and the mechanism of the self-change of the cave1-3 in the endurance exercise is discussed. And provides a theoretical basis for the change of the cell structure caused by the endurance exercise and the induced fatigue rapid recovery. CsA and endurance Under the influence of exercise, the change of NOS was not obvious and the content of NO increased.4. CsA intervention in that next group, the LDH content in the tissue increase and the CK change is restored to a quiet time . LDH recovery is slower than CK, and the time to reflect the damage is longer.5. Long-term endurance exercise, CsA intervention and the synergistic effect of both can cause Caveolin in the muscle of different fiber types. the reduction of the content of -3, affecting the signaling pathway mediated by the Caveolin-3.
