放射性肺损伤早期的肺组织重建机制研究

目的:用分子生物学和细胞生物学方法,从细胞水平和分子水平深入研究肺纤维化的发生机制,研究肺泡巨噬细胞、成纤维细胞、Ⅳ型胶原和明胶酶B在放射性肺损伤中的关系,进而阐明肺组织重建的机理及其与肺纤维化发生之间的联系。 方法:用一定剂量的60Coγ射线照射C57小鼠和Wistar大鼠。将C57小鼠分为2组:照射组(n=9)和对照组(n=9)。分别于照后1、2、4w活杀取其肺组织提取总RNA。将Wistar大鼠分为对照组和照射后7d组,用0.9%的生理盐水灌洗肺脏,用差速贴壁法分离肺泡巨噬细胞,并用台盼蓝排斥试验鉴定其存活率>95%,然后用分离好的巨噬细胞制备肺泡巨噬细胞条件培养液(Condition Medium of Alveolar Macrophage,CMAM),用于下一步试验。MTT法检测CMAM对人肺成纤维细胞(Human Lung Fibroblast,HLF)的促增殖作用,于不同的条件培养液培养HLF 6h、12h、24h、48h、72h后检测HLF的增殖情况。用CMAM大量培养HLF,于培养后的不同时间点提取总蛋白,分别用Western Blot和...
