海水对兔眼晶状体功能及形态学影响的实验研究

【摘要】:目的:观察将海水注入前房对兔眼晶状体功能及形态学的影响,并对其致伤机制进行探讨。 方法:随机选择55只兔子中的50只建立动物模型,右眼前房注入0.2ml海水,分别于术后15分钟、30分钟、1小时、3小时、6小时及12小时经裂隙灯显微镜检查后摘除眼球行光电镜检查及Na~+,K~+-ATP酶、Ca~(2+),Mg~(2+)-ATP酶、琥珀酸脱氢酶(Succinate dehydrogenase,SDH)、乳酸脱氢酶(Lactate dehydrogenase,LDH)、超氧化物歧化酶(Superoxide Dismutase,SOD)和谷光甘肽过氧化物酶(Glutathione-Peroxide,GSH-PX)的生化活性检测。左眼前房内注入0.2ml平衡盐溶液(Balanced salt solution,BSS)作为自身对照,其余步骤同右眼,另外5只兔子不做任何处理为正常组。 结果:1、裂隙灯显微镜观察海水组兔子晶状体起初明显混浊,6小时后混浊晶状体逐渐恢复透明。2、光镜下海水组细胞表现为水肿、变性、坏死及Morgagnian小体形成,对照组细胞轻度水肿,无明显Morgagnian小体形成。3、电镜下海水组细胞逐渐出现线粒体、粗面内质网和高尔基复合体水肿扩张及空泡化改变,对照组细胞内超微结构轻度水肿。4、海水组品状体Na~+,K~+-ATPase及Ca~(2+),Mg~(2+)-ATPase活性均先降低,再增高,最后完全或部分恢复正常;SDH及LDH活性6小时内均代偿性增高,12小时恢复正常;SOD
[Abstract]:Aim: to observe the effects of seawater injection into anterior chamber on lens function and morphology of rabbit eyes, and to explore the mechanism of injury. Methods: fifty of 55 rabbits were randomly selected to establish animal models, and 0.2ml seawater was injected into the right anterior chamber. After 15 minutes, 30 minutes, 1 hour, 3 hours, 6 hours and 12 hours, the eyeballs were removed by slit lamp microscope and examined with photoelectricity microscope and Na ~ + K ~ -ATPase. The biochemical activities of succinate dehydrogenase (Succinate dehydrogenase), lactate dehydrogenase (Lactate dehydrogenase), superoxide dismutase (SOD) and glutathione-peroxidase (Glutathione-Peroxide-GSH-PX) were detected. 0.2ml balanced salt solution was injected into the left anterior chamber as self-control, the other steps were the same as the right eye, and the other 5 rabbits were not treated as normal group. Results under the slit lamp microscope, the lens of rabbits in seawater group recovered gradually to hyaline 0.2 after 6 hours of obvious opacification. The cells of seawater group showed edema, degeneration, necrosis and formation of Morgagnian bodies under light microscope, while those in control group showed mild edema. There was no obvious formation of Morgagnian corpuscles. Under electron microscope, mitochondria, rough endoplasmic reticulum and Golgi complex were gradually appeared in the cells of seawater group. In the control group, the ultrastructure of the cells was slightly edematous .4.The activities of Na ~ + K ~ -ATPase and Ca~(2 _ 2 ~ -ATPase decreased first, then increased, and finally returned to normal completely or partially within 6 hours, and then returned to normal in 12 hours.
