晕船及其适应过程中大鼠氨基酸代谢变化研究

【摘要】:目的观察模拟晕船及晕船适应后大鼠血浆及组织内游离氨基酸含量变化,为进一步研究防治晕船提供实验依据。方法:采用Crampton模型模拟晕船刺激,以异嗜高岭土行为作为判定大鼠晕船及晕船适应的指标,运用高效液相色谱法测定晕船及晕船适应过程中大鼠血浆及组织内游离氨基酸含量变化。结果:模拟晕船刺激1d后,血浆Cys和Ile含量显著升高,Gly、Pro含量显著下降,BCAA/AAA的比值升高;脑内Ala,Cys+Met,Tyr,His以及TAA含量显著下降;肝脏内Ser、Val、Ile、Leu、Phe、Lys含量显著升高,His含量显著下降,AAA/BCAA的比值降低;肾内Ser、Ala、Phe含量下降。模拟晕船刺激21d后脑内Glu/GABA显著升高。肾EAA含量和EAA/TAA的比值显著降低。结论:晕船刺激使实验大鼠血浆和组织内氨基酸含量发生明显改变,其中有些与神经递质合成有关的氨基酸改变较显著。
[Abstract]:Objective to observe the changes of free amino acids in plasma and tissue after simulated seasickness and seasickness adaptation in rats, and to provide experimental evidence for further study on prevention and treatment of seasickness. Methods: Crampton model was used to simulate seasickness stimulation and isophilic kaolin behavior was used as an index to judge seasickness and seasickness adaptation in rats. The contents of free amino acids in plasma and tissue of seasickness and seasickness adaptation rats were determined by high performance liquid chromatography (HPLC). Results: after stimulated by simulated seasickness for 1 day, the contents of Cys and Ile in plasma increased significantly, the content of Gly,Pro decreased, the ratio of BCAA/AAA increased, and the contents of Ala,Cys Met,Tyr,His and TAA in brain decreased significantly. The content of Ser,Val,Ile,Leu,Phe,Lys in liver increased significantly, the content of His decreased significantly, the ratio of AAA/BCAA decreased, and the content of Ser,Ala,Phe in kidney decreased. Glu/GABA in the brain increased significantly after simulated seasickness stimulation for 21 days. The ratio of EAA content to EAA/TAA in kidney was significantly decreased. Conclusion: the contents of amino acids in plasma and tissue of experimental rats were significantly changed by seasickness stimulation, some of which were related to neurotransmitter synthesis.
