【摘要】:针灸疗法是中医学体系的一个重要组成部分,对一些疾病(例如慢性疼痛)具有良好的治疗效果。针灸效应作用机制至今尚未解决。研究针灸效应的细胞响应有助于理解针灸效应机理,推动针灸经络相关研究,促进传统中医学的发展。 本文采用光学检测技术研究针灸效应在细胞水平的生理表征。首先,利用激光扫描共聚焦显微成像技术检测了活细胞中针灸效应作用分子一氧化氮的含量,并对比了不同类型细胞中一氧化氮含量的差异。其次,研究了一氧化氮对Hela细胞内钙离子浓度的调节作用。研究表明一氧化氮能诱导细胞内钙离子浓度升高,而且一氧化氮分子诱导钙离子浓度升高是细胞外钙离子内流和细胞内钙库释放钙离子共同作用的结果。内部钙库释放钙离子是钙离子浓度升高的主要影响因素。研究还表明线粒体是一氧化氮分子引起细胞内部释放钙离子的一种钙库。再次,基于生物发光共振能量转移技术研究了腺苷在激光针灸效应中的作用。研究发现激光辐照能抑制腺苷含量,且抑制程度与辐照光剂量呈正比。此外,腺苷A3受体拮抗剂能增强激光辐照对腺苷的抑制作用。最后,利用数学模型模拟了激光辐照对动作电位的影响。结果表明激光辐照能缩小动作电位的周期,激光辐照剂量对动作电位传输特性有一定的影响。较高辐照光剂量能够抑制动作电位的传输,而较小辐照光剂量能促进动作电位的传输。 本文探索了针灸效应的若干细胞响应,研究结果有助于在细胞分子水平认识针灸效应的相关机理。
[Abstract]:Acupuncture and moxibustion therapy is an important part of traditional Chinese medicine system. The mechanism of acupuncture and moxibustion effect has not been solved yet. The study of cellular response of acupuncture and moxibustion effect is helpful to understand the mechanism of acupuncture effect. To promote the research of acupuncture and moxibustion meridian and promote the development of traditional Chinese medicine. In this paper, the physiological characterization of acupuncture and moxibustion effect at cell level was studied by optical detection. Firstly, the content of nitric oxide (no) in living cells was detected by laser scanning confocal microscopy (LSCM). The differences of nitric oxide content in different types of cells were compared. The effect of nitric oxide on intracellular calcium concentration in Hela cells was studied. Moreover, the increase of calcium concentration induced by nitric oxide is the result of the interaction between extracellular calcium influx and intracellular calcium release, and the internal calcium release is the main influencing factor for the increase of calcium concentration. Studies have also shown that mitochondria are a calcium pool that is released by nitric oxide (no) molecules inside cells. The effects of adenosine on the effect of laser acupuncture and moxibustion were studied based on the bioluminescence resonance energy transfer technique. It was found that laser irradiation could inhibit the content of adenosine, and the degree of inhibition was proportional to the irradiation dose. Adenosine A3 receptor antagonist can enhance the inhibitory effect of laser irradiation on adenosine. Finally, the effect of laser irradiation on action potential is simulated by mathematical model. The results show that laser irradiation can reduce the period of action potential. The higher irradiation dose can inhibit the action potential transmission, while the lower irradiation light dose can promote the action potential transmission. Several cellular responses of acupuncture and moxibustion effects are explored in this paper. The results are helpful to understand the mechanism of acupuncture effect at cellular and molecular level.
