电针对淀粉样前体蛋白转基因小鼠海马微血管淀粉样沉积的影响及其与低密度脂蛋白相关受体1的关系

【摘要】:目的:探讨电针治疗阿尔茨海默病的可能作用机制。方法:将13月龄的淀粉样前体蛋白(APP)转基因小鼠随机分为模型组和电针治疗组,以相同月龄和背景的C 57 BL/6小鼠作对照组。电针治疗组取"百会""涌泉"穴,每次留针15 min,隔日1次,针刺3个月。以LashleyⅢ水迷宫测定各组小鼠的学习记忆能力,以免疫组化方法检测海马β淀粉样蛋白(amyloidβprotein,Aβ)、低密度脂蛋白相关受体1(low densi-ty lipoprotein receptor-related protein 1,LRP 1)的表达。结果:模型组水迷宫游出时间较对照组延长(P0.05),海马沟微血管壁Aβ1-42表达的累积吸光度较对照组升高(P0.01),海马沟微血管壁LRP 1表达较对照组减低(P0.01);而电针治疗组的水迷宫游出时间明显低于模型组,海马沟微血管壁Aβ1-42表达低于模型组,海马沟微血管壁LRP 1表达高于模型组(均P0.05)。结论:电针治疗可能通过提高脑微血管壁Aβ受体LRP 1的清除转运能力,降低APP转基因鼠脑微血管壁的Aβ沉积,从而改善其学习、记忆能力。
[Abstract]:Objective: to explore the possible mechanism of electroacupuncture in the treatment of Alzheimer's disease. Methods: the 13 month old transgenic mice with amyloid precursor protein (APP) were randomly divided into two groups: model group and electroacupuncture group.
