针刺对偏头痛大鼠血清SP、CGRP影响的实验研究

【摘要】:目的:通过复制皮下注射硝酸甘油偏头痛大鼠模型,,观察针刺对偏头痛大鼠行为学变化的影响及对血清内SP、CGRP含量变化的影响,探讨针刺预防及治疗偏头痛的机制。 材料与方法: 1.健康雄性SD大鼠50只,体重约为250+50g,适应性喂养一周之后开始实验。按照随机数字表法将大鼠分为5组(空白对照组、模型对照组、针刺预防组、针刺治疗组、针刺预防+治疗组),每组10只。空白对照组:正常饲养7天,不进行任何处理;模型对照组:正常饲养7天,第8天皮下注射硝酸甘油造模;针刺预防组:针刺治疗7天,20min/d,第8天皮下注射硝酸甘油造模;针刺治疗组:正常饲养7天,第8天皮下注射硝酸甘油造模,造模成功后,针刺20min一次;针刺预防+治疗组:针刺治疗7天,20min/d,第8天皮下注射硝酸甘油造模,造模成功后,针刺20min一次。 2.皮下注射硝酸甘油(10mg/kg)制备偏头痛大鼠模型。 3.针刺取穴:百会、阳陵泉(双侧)、太冲(双侧)、外关(双侧)。 4.观察造模后大鼠行为学变化,采用以下三项作为观察指标:①大鼠耳红持续时间:观察并记录大鼠双耳发红出现和消失的时间点并计算其持续时间;②大鼠前肢频繁挠头行为持续时间:观察并记录大鼠前肢频繁挠头(以连续5次以上挠头为标准)行为出现时间点及消失(连续挠头次数不及5次)时间点,并计算其持续时间;③分时间段统计大鼠挠头次数:以30分钟为一时间段,分时间段记录自造模31分钟后至180分钟内挠头总次数。 5.采用酶联免疫吸附法(ELISA)测定大鼠血清中SP、CGRP的含量。 结果: 1.模型对照组在皮下注射硝酸甘油后大概4min左右即出现耳红、前肢频繁挠头、症状,此症状持续170-180分钟不等。各组与同时间段空白对照组比较显示在各个时间段内均有前肢频繁挠头次数增多症状(P0.05)说明造模成功。 2.针刺对大鼠行为学变化的影响:对耳红、前肢频繁挠头持续时间进行统计可发现针刺预防组、针刺治疗组、针刺预防+治疗组均能有效降低耳红、前肢频繁挠头症状的持续时间,与模型对照组比较有统计学意义(P0.05);以30min为一时间段,分时间段内统计31-180min内大鼠频繁挠头次数发现,针刺预防组、针刺治疗组、针刺预防+治疗组在各个时间段内均能有效降低挠头次数,与同时间段内模型对照组比较P0.05,针刺预防+治疗组与针刺预防组、针刺治疗组比较疗效更佳(P0.05)。 3.针刺对大鼠血清内SP、CGRP含量变化的影响:针刺预防组与模型对照组比较:SP、CGRP含量降低,证明针刺对偏头痛大鼠有预防作用(P0.05);针刺治疗组与模型对照组比较:SP、CGRP含量降低,证明针刺对偏头痛大鼠有治疗作用(P0.05);针刺预防+治疗组与针刺预防组、针刺治疗组比较:SP、CGRP含量下降,证明针刺预防+治疗组的疗效优于单纯治疗及单纯预防(P0.05)。 结论: 1参照Cristina方法,成功复制出硝酸甘油型偏头痛大鼠模型。 2.针刺可有效改善偏头痛大鼠行为学变化,以针刺预防+治疗组疗效明显。 3.硝酸甘油型偏头痛大鼠模型血清内SP、CGRP含量升高,推测偏头痛的发生与SP介导的超敏反应与CGRP引起的血管过度扩张及神经源性炎症有关。 4.针刺可有效降低偏头痛大鼠血清内SP、CGRP含量,以针刺预防+治疗组效果明显。 5.针刺可能通过降低偏头痛大鼠血清内SP、CGRP的含量,进而缓解脑血管过度扩张及超敏反应,调节脑血流量,减轻神经源性炎症,来对偏头痛产生预防及治疗作用。
[Abstract]:Objective: To observe the effect of acupuncture on the behavioral changes of migraine rats and the effect of acupuncture on the changes of serum SP and CGRP contents, and to explore the mechanism of acupuncture prevention and treatment of migraine by replicating the model of migraine with subcutaneous injection of nitroglycerin.
Materials and methods:
1. of 50 healthy adult male SD rats, weighing about 250+50g, fed for one week after the start of the experiment. According to the random number table method, the rats were divided into 5 groups (blank control group, model control group, acupuncture prevention group, acupuncture group, acupuncture prevention + treatment group), 10 rats in each group. The blank control group: normal feeding for 7 days without any treatment; model control group: normal feeding for 7 days, eighth days subcutaneous injection of nitroglycerin model; acupuncture prevention group: acupuncture treatment for 7 days, 20min/d eighth days subcutaneous injection of nitroglycerin model; acupuncture treatment group: normal feeding for 7 days, eighth days subcutaneous injection Nitroglycerol., after the success of modeling, 20min acupuncture once; acupuncture prevention + treatment group: acupuncture treatment for 7 days, 20min/d eighth days, subcutaneous injection of Nitroglycerol., after the success of the model, acupuncture 20min once.
2. the rat model of migraine was prepared by subcutaneous injection of nitroglycerin (10mg/kg).
3. acupoints: Baihui, Yanglingquan (bilateral), Taichong (bilateral), Waiguan (bilateral).
4. to observe the behavior changes of rats after modeling, using the following three items as observation index: rat ear red duration: observe and record the rat red ears appear and disappear time points and calculate the duration; the rat forelimb duration: frequent scratching behavior were observed and recorded rat forelimb frequent scratching (for more than 5 consecutive head as the standard) appeared time and disappeared (continuous scratching number is less than 5 times) time point, and calculate the duration; the time statistics of rats: scratching frequency of 30 minutes for a period of time, the time period of record from the model after 31 minutes to 180 minutes. The total number of scratching.
5. the content of SP and CGRP in the serum of rats was determined by enzyme linked immunosorbent assay (ELISA).
Result:
The 1. model control group in the subcutaneous injection of nitroglycerin after about 4min appears red ear, forelimb frequent scratching, symptoms, the symptoms lasted 170-180 minutes. From the same time group and blank control group shows in different periods have increased the frequency of symptoms of forelimb frequent scratching (P0.05) indicated that the model was successful.
2. effect of acupuncture on rat behavior change: the red ear, frequent scratching duration of forelimb statistics can be found in acupuncture prevention group, acupuncture group, acupuncture prevention + treatment group can effectively reduce ear red, duration of symptoms and forelimb frequent scratching, compared with the model control group was statistically significant (P0.05) for a period of time; in 30min, divided time statistics 31-180min rats found frequent scratching frequency, acupuncture prevention group, acupuncture group, acupuncture prevention + treatment group can effectively reduce the scratching frequency in each time, with the same period in the model control group P0.05 prevention + treatment group and Acupuncture Acupuncture prevention group, acupuncture treatment group has better curative effect (P0.05).
3. acupuncture on rats serum SP, CGRP content effects: Acupuncture prevention group compared with the control group: SP, CGRP content decreased, that acupuncture has a preventive effect on migraine rats (P0.05); acupuncture treatment group compared with the control group: SP, CGRP content decreased, evidence of acupuncture therapy in migraine rats (P0.05); acupuncture prevention + treatment group and acupuncture prevention group, acupuncture treatment group comparison: SP, CGRP content decreased, showed that acupuncture efficacy in the prevention + treatment group was superior to that of simple treatment and Prevention (P0.05).
Conclusion:
1 the nitroglycerin type migraine rat model was successfully replicated with reference to the Cristina method.
2. acupuncture can effectively improve the behavioral changes of migraine rats, and the curative effect of acupuncture prevention + treatment group is obvious.
3. the nitroglycerin type migraine rat model increased serum SP and CGRP levels. It is speculated that the occurrence of migraine and SP mediated hypersensitivity are related to vascular overexpansion and neurogenic inflammation caused by CGRP.
4. acupuncture can effectively reduce the content of SP and CGRP in the serum of migraine rats, and the effect of acupuncture prevention + treatment group is obvious.
5. acupuncture can reduce the content of SP and CGRP in the serum of migraine rats, relieve cerebral vascular overexpansion and hypersensitivity reaction, regulate cerebral blood flow and reduce neurogenic inflammation, so as to prevent and treat migraine.
