中西医结合治疗原发性胆汁性肝硬化小鼠疗效分析

【摘要】:探讨熊去氧胆酸(UDCA)和"一贯煎"在原发性胆汁性肝硬化小鼠中的疗效。对polyI∶C注射的C57BL/6雌性小鼠分别给予UDCA、"一贯煎"和两种药物联合治疗,分析不同药物治疗组血清ALP、-γGT的变化,实时荧光定量RT-PCR方法检测脾脏单个核细胞BCMA mRNA的表达,并分析肝门管区单个核细胞浸润率。结果表明,PBC模型鼠肝门管区单个核细胞浸润率、脾脏单个核细胞BCMA mRNA的表达较正常小鼠明显增加(P0.05);不同治疗组较模型对照组ALP、-γGT显著下降(P0.05),"一贯煎"或联合治疗还能减少肝门管区单个核细胞浸润率、下调脾脏单个核细胞BCMAmRNA和ALP。"一贯煎"或联合熊去氧胆酸治疗PBC小鼠疗效更好,为以后中西医联合治疗PBC提供了理论依据和技术支持。
[Abstract]:To investigate the efficacy of ursodeoxycholic acid (UDCA) and "Yijian" in primary biliary cirrhosis mice. The female C57BL/6 mice injected with polyI:C were treated with UDCA, "Yijian" and two drugs, respectively. The changes of serum ALP- 纬 GT in different drug treatment groups were analyzed, and the expression of BCMA mRNA in spleen mononuclear cells was detected by real-time fluorescence quantitative RT-PCR. The infiltration rate of mononuclear cells in hepatic hilar area was analyzed. The results showed that the infiltration rate of mononuclear cells in hepatic hilar area of PBC model rats, The expression of BCMA mRNA in splenic mononuclear cells was significantly higher than that in normal mice, and the expression of BCMA mRNA in spleen mononuclear cells was significantly decreased in different treatment groups than that in the model control group. "Yijianjian" or combined treatment could also reduce the infiltration rate of mononuclear cells in the hilar area of the liver and down-regulate the BCMAmRNA and ALP of splenic mononuclear cells. " Yijianjian "or ursodeoxycholic acid combined with ursodeoxycholic acid is more effective in treating PBC mice, which provides theoretical basis and technical support for the later treatment of PBC by combination of traditional Chinese and western medicine."
