【摘要】:肿瘤与细胞的异常分化直接相关。胚胎发育,是细胞不断增殖、诱导正常细胞分化、并伴有凋亡的过程,而在这其间出现的异常细胞分化则会被诱导修正。胚胎在发育过程中能够产生促分化因子,诱导胚胎细胞分化、凋亡,决定胚胎发育的方向。近年来促分化因了的应用,尤其在诱导肿瘤细胞凋亡方面,引起了人们的广泛关注。经本实验室前期研究发现,动物的胚胎蛋白在诱导肿瘤细胞凋亡方面作用显著,并且具有普遍性。本文进一步验证,胚胎提取蛋白对于多种肿瘤细胞的体外培养过程中,均具有抑制细胞增殖、诱导凋亡的作用,并初步验证胚胎蛋白对小鼠体内肿瘤细胞同样具有诱导凋亡的作用。本文选用鸡胚胎提取蛋白以及蛙胚胎提取蛋白作为诱导物,以不同浓度梯度分别诱导多同肿瘤细胞,以比对其诱导凋亡差异性。选用的肿瘤细胞主要有:RM1、HepG2、NLT、MCF7、Hela、B16等。通过光学显微镜、电子显微镜、激光共聚焦显微镜,以及MTT,流式细胞仪等检测,证实胚胎蛋白对各种肿瘤细胞均具有抑制增殖、诱导凋亡的效果,且具有诱导物浓度影响诱导效果的差异。将B16细胞接种到小鼠皮下,建立体内肿瘤动物模型,采用原位注射法,将胚胎蛋白注入小鼠体内,结果证实,在实验组小鼠体内,肿瘤的体积,与对照组相比明显减小。初步验证了胚胎蛋白对生长在小鼠体内的肿瘤细胞同样具有诱导凋亡的作用。
[Abstract]:Tumor is directly related to abnormal differentiation of cells. Embryonic development is the process of cell proliferation, induction of normal cell differentiation and apoptosis. However, the abnormal cell differentiation will be induced and corrected during the process of embryo development, which can induce differentiation and apoptosis of embryonic cells. In recent years, the application of differentiation promotion, especially in inducing apoptosis of tumor cells, has attracted wide attention. Animal embryonic proteins play a significant role in inducing apoptosis of tumor cells, and they are universal. In this paper, we further verify that embryonic proteins can be used in the culture of various tumor cells in vitro. Both of them can inhibit cell proliferation and induce apoptosis. It was also proved that embryonic protein could induce apoptosis in mouse tumor cells. Chicken embryo extract protein and frog embryo extract protein were selected as inducers. Different concentrations of gradient were used to induce the same tumor cells, and to compare the difference of inducing apoptosis. The main tumor cells selected were: RM1, HepG2, NLTG2NLTMCF7 and Hela. B16 et al. By optical microscope, electron microscope, laser confocal microscope, MTT, flow cytometry and other detection, it was proved that embryonic protein can inhibit the proliferation of various tumor cells. B16 cells were inoculated subcutaneously into mice to establish tumor animal model in vivo, and in situ injection method was used. Embryonic proteins were injected into mice and the results showed that the tumor volume was measured in mice of experimental group. Compared with the control group, it was proved that embryonic protein could induce apoptosis of tumor cells in mice.
