低剂量辐射通过下调ERCC1和Bcl-2增强耐药卵巢癌细胞对顺铂的敏感性

【摘要】:目的:卵巢癌是所有女性恶性肿瘤中容易引起死亡的疾病。而卵巢癌的主要治疗方法是手术治疗加上术后的多药联合化疗,其中铂类是多药联合化疗中的基础药物。然而对于卵巢癌来说,其主要的挑战来自癌细胞对于化疗药物抵抗性的发生。所以本试验的目的在于证实低剂量辐射有增强耐药卵巢癌细胞化疗敏感性的作用,并探究其机制,然后为耐药性卵巢癌的临床治疗提供新的策略。方法:CCK8法检测细胞增殖。采用流式细胞仪(FCM)的方法,通过Annexin V-FITC和PI的双染色来量化耐顺铂卵巢癌细胞(SKOV3/DDP)的凋亡率。实时荧光定量PCR(q PCR)分析SKOV3/DDP细胞中ERCC1和Bcl-2的m RNA表达水平。结果:对照组,常规剂量组和低剂量组的IC50值分别为9.367±0.16,9.289±0.16和3.847±0.15分别(P0.05)。经过流式细胞仪检测,低剂量辐射组较其他组具有更显著的凋亡率(p0.05)。低剂量组的ERCC1和Bcl-2的m RNA相对表达量比对照组及常规剂量组显著降低(p0.05)。结论:低剂量辐射具有增加SKOV3/DDP细胞对化疗药物(顺铂)敏感性的作用,而其这一作用可能是通过减弱SKOV3/DDP细胞的DNA损伤修复能力和加强肿瘤细胞的凋亡来实现的。
[Abstract]:Objective: ovarian cancer is one of the leading causes of death in all female malignancies. The main treatment for ovarian cancer is surgical treatment plus postoperative multidrug chemotherapy. Among them, platinum is the basic drug in multidrug combination chemotherapy. However, for ovarian cancer. The main challenge is the development of cancer cell resistance to chemotherapeutic drugs, so the aim of this study is to confirm that low dose radiation enhances chemosensitivity of drug-resistant ovarian cancer cells and to explore its mechanism. Then it provided a new strategy for the clinical treatment of drug-resistant ovarian cancer. Methods the cell proliferation was detected by the method of: CCK8. Flow cytometry (FCM) was used. The apoptosis rate of cisplatin resistant ovarian cancer cell line SKOV3 / DDP was quantified by double staining of Annexin V-FITC and Pi. Real-time fluorescence quantitative PCR(q). The expression of m RNA in ERCC1 and Bcl-2 in SKOV3/DDP cells was analyzed. Results: control group. The IC50 values of routine and low dose groups were 9.367 卤0.16, respectively. 9.289 卤0.16 and 3.847 卤0.15, respectively. The relative expression of m RNA in ERCC1 and Bcl-2 in the low dose group was significantly lower than that in the control group and the normal dose group (P < 0. 05), and the relative expression of m RNA in the low dose group was significantly lower than that in the control group and the normal dose group (P < 0. 05). Conclusion: low dose radiation can increase the sensitivity of SKOV3/DDP cells to chemotherapeutic drugs (cisplatin). This effect may be achieved by weakening the DNA damage and repair ability of SKOV3/DDP cells and enhancing the apoptosis of tumor cells.
