miR-135b对子宫内膜癌HOXA10基因表达的调控作用

【摘要】:目的探讨miR-135b对子宫内膜癌中HOXA10基因表达的调控作用。方法 RT-PCR方法检测28例子宫内膜癌组织及相应癌旁组织中miR-135b及HOXA10的表达;用miR-135b模拟物及抑制物转染RL-952子宫内膜癌细胞株,RT-PCR检测转染效果并检测FOXO1 mRNA的变化,划痕实验检测转染后对细胞迁移的影响,流式细胞技术检测转染对细胞增殖的影响,Western blot检测HOXA10蛋白的变化。结果 miR-135b在子宫内膜癌组织中较对应的癌旁组织表达升高(P0.05),HOXA10则降低(P0.05),经miR-135b模拟物转染后,子宫内膜癌细胞中HOXA10 mRNA和蛋白的表达水平下降(P0.05);经miR-135b抑制物转染后,子宫内膜癌细胞中HOXA10 mRNA和蛋白的表达水平升高(P0.05);miR-135b能促进子宫内膜癌细胞的增殖(P0.05)。结论 miR-135b在子宫内膜癌发生发展中有一定作用,子宫内膜中HOXA10的异常表达受miR-135b的调控。
[Abstract]:Objective to investigate the effect of miR-135b on the expression of HOXA10 gene in endometrial carcinoma. RT-PCR method was used to detect the expression of miR-135b and HOXA10 in 28 cases of endometrial carcinoma and its adjacent tissues. The transfection effect and FOXO1 mRNA of RL-952 endometrial carcinoma cell line were detected by RT-PCR with miR-135b mimic and inhibitor. The effect of transfection on cell migration was detected by scratch test, and the effect of transfection on cell proliferation was detected by flow cytometry. Western blot was used to detect the changes of HOXA10 protein. Results the expression of miR-135b in endometrial carcinoma tissues was higher than that in the corresponding adjacent tissues (P0.05). The expression of HOXA10 mRNA and protein in endometrial cancer cells decreased after transfection with miR-135b mimics. After transfection with miR-135b inhibitor, the expression of HOXA10 mRNA and protein increased in endometrial carcinoma cells. MiR-135b can promote the proliferation of endometrial carcinoma cells. Conclusion miR-135b may play a role in the development of endometrial carcinoma. Abnormal expression of HOXA10 in endometrium is regulated by miR-135b.
