整合素α5和caspase 3在肝癌中的表达及其临床意义

【摘要】：目的研究整合素α5(integrinα5,ITGA5)和caspase 3在肝癌组织中的表达及其临床意义。方法1.免疫组化Elivision二步法检测ITGA5和caspase 3在36例肝癌组织石蜡标本、23例癌旁组织石蜡标本中的表达。分析ITGA5、caspase 3在肝癌组织中的表达与临床病理特征的关系。2.荧光定量RT-PCR、Western blot分别检测经外科手术切除冻存的25例肝细胞癌患者的癌组织及癌旁组织中ITGA5和caspase 3 mRNA及蛋白质的相对表达量。3.SPSS20.0统计软件用于分析实验数据,计量资料以ⅹ_结果1.免疫组化染色结果±s表示,两组间比较采用独立样本t检验,计数资料比较采用卡方或Fisher精确概率法,两变量关联性采用Spearman相关性分析,以P0.05为差异有统计学意义。1.1 ITGA5蛋白质主要表达于细胞膜和细胞质,其在肝癌组织和癌旁组织中的阳性表达率分别为88.89%(32/36),43.48%(10/23),差异有统计学意义(χ2=14.109,P0.01)。ITGA5在肝癌中的表达与TNM分期及肿瘤转移有关(P均0.05)。TNM分期为Ⅲ~Ⅳ期的肝癌组织ITGA5阳性表达率更高;伴有转移的肝癌组织ITGA5阳性表达率更高。1.2 caspase 3蛋白质主要表达于细胞质,其在肝癌组织和癌旁组织中的阳性表达率分别为30.56%(11/36),86.96%(20/23),差异有统计学意义(χ2=17.903,P均0.01)。caspase 3在肝癌中的表达与HBsAg、TNM分期及肿瘤转移有关(P均0.05)。HBsAg阳性、TNM分期为Ⅲ~Ⅳ期、伴有转移的肝癌组织caspase 3阳性表达率更低。2.荧光定量RT-PCR检测结果显示:ITGA5 mRNA在肝癌组织中的相对表达量(3.188±1.339)高于癌旁组织中的相对表达量(0.388±0.238);caspase 3 mRNA在肝癌组织中的相对表达量(0.209±0.108)低于癌旁组织中的相对表达量(5.755±2.522)。差异均有统计学意义(P均0.01)。3.Western blot检测结果显示:ITGA5蛋白质在肝癌组织中的相对表达量(0.583±0.041)高于癌旁组织中的相对表达量(0.048±0.029);caspase 3蛋白质在肝癌组织中的相对表达量(0.168±0.072)低于癌旁组织中的表达量(3.885±0.629)。差异均有统计学意义(P均0.01)。4.肝癌组织中,ITGA5与caspase 3的表达呈负相关(r=-0.341,P0.05)。结论ITGA5在肝癌组织中表达增高,且和caspase 3的表达呈负相关,并与肝癌的TNM分期及肿瘤转移有关,提示其表达的高低可能与肝癌转移有关。
