ADAM8基因在胃癌组织中的表达及与临床病理特征的关系

【摘要】：目的检测ADAM8基因在胃癌组织中的表达水平,探讨胃癌的生物学特征与其表达之间的关系。方法应用免疫组织化学、实时定量PCR及Western blot技术检测100例胃癌组织、20例正常胃黏膜组织中ADAM8基因的表达,并分析其与临床病理特征的相关性。结果免疫组织化学分析显示ADAM8在正常胃黏膜组织中表达率为30%(6/20),而胃癌组织中ADAM8的表达率为86%(86/100),提示ADAM8基因表达在胃癌组织中显著上调(P0.05);实时定量PCR检测显示在胃癌标本中ADAM8 mRNA的表达水平为(0.767±0.129),在正常胃组织中表达水平为(0.192±0.127),两者差异有统计学意义(P0.01)。Western blot检测结果显示在胃癌组织中ADAM8蛋白表达水平为(0.659±0.027),而正常胃黏膜标本中其表达水平为(0.236±0.074),差异有统计学意义(P0.01)。且伴淋巴结转移胃癌组织、无淋巴结转移胃癌组织与正常胃黏膜组织相比,ADAM8蛋白表达水平明显增高,差异均具有统计学意义(均P0.05)。进一步分析ADAM8蛋白表达与临床病理特征之间的关系,结果显示其表达与肿瘤的直径、临床分期、肿瘤的浸润深度、有无转移等显著相关(均P0.05);与性别、年龄等因素无明显相关(均P0.05)。结论 ADAM8在胃癌组织中的高表达与胃癌侵袭转移有关,提示ADAM8可能参与胃癌的发生、发展过程,可能成为胃癌预后判断的参考指标。
[Abstract]:Objective to detect the expression level of ADAM8 gene in gastric carcinoma and to explore the relationship between its biological characteristics and its expression. Methods Immunohistochemistry was used. Real-time quantitative PCR and Western blot techniques were used to detect the expression of ADAM8 gene in 20 normal gastric mucosa tissues. Results Immunohistochemical analysis showed that the expression rate of ADAM8 in normal gastric mucosa was 30 / 6 / 20). The expression rate of ADAM8 in gastric cancer was 86 / 100, suggesting that the expression of ADAM8 gene was significantly up-regulated in gastric cancer tissues. The expression level of ADAM8 mRNA in gastric cancer was 0.767 卤0.129 by real-time quantitative PCR. The expression level in normal gastric tissues was 0.192 卤0.127). The expression level of ADAM8 protein in gastric carcinoma was 0.659 卤0.027). The expression level in normal gastric mucosa was 0.236 卤0.074, the difference was statistically significant (P 0.01). The expression of ADAM8 protein in gastric cancer without lymph node metastasis was significantly higher than that in normal gastric mucosa. The relationship between the expression of ADAM8 protein and the clinicopathological features was further analyzed. The results showed that the expression of ADAM8 protein was related to the diameter and clinical stage of the tumor. The depth of invasion and metastasis were significantly correlated (all P 0.05); Conclusion the high expression of ADAM8 in gastric cancer is related to invasion and metastasis of gastric cancer, suggesting that ADAM8 may be involved in the occurrence of gastric cancer. The process of development may be a reference index for judging the prognosis of gastric cancer.
