血根碱对舌癌体外增殖、凋亡和迁移作用的影响

【摘要】:目的:研究博落回提取物中血根碱(Sanguinarine,SAN)对舌癌体外增殖、凋亡和迁移作用的影响及其机制。方法:采用不同浓度的血根碱处理Tscca和Tca8113细胞24、48、72 h,MTT法检测血根碱对两种舌癌细胞增殖抑制的影响;用药48 h后,流式细胞仪Annexin V-FITC/PI染色法检测不同浓度血根碱对细胞凋亡的影响;Western blotting法检测不同浓度血根碱对细胞中NF-κB表达的影响;细胞划痕实验检测不同浓度血根碱对舌癌细胞迁移能力的影响。结果:经血根碱处理后的舌癌细胞系Tscca和Tca8113有明显的抑制增殖作用,并呈现一定的浓度和时间依赖关系。血根碱作用于舌鳞癌Tca8113细胞24 h、48 h、72h的半数抑制浓度(IC50)分别为2.53μmol/L、2.38μmol/L、1.96μmol/L。舌鳞癌Tscca细胞24 h、48 h、72h的半数抑制浓度(IC_(50))分别为1.48μmol/L、1.17μmol/L、0.89μmol/L。血根碱可以抑制NF-κB的表达,且有一定的剂量相关性。一定浓度的血根碱还能抑制Tca8113细胞株的迁移。结论:血根碱能显著抑制舌癌细胞Tscca和Tca8113的增殖并且诱导其凋亡,其机制可能与对NF-κB表达的影响有关,同时血根碱可影响Tca8113细胞株的迁移。
[Abstract]:Objective: to study the effect of Sanguinarine (SAN) on the proliferation of tongue cancer in vitro. Methods: Tscca and Tca8113 cells were treated with different concentrations of hematopoietic alkaloids for 24 872 h, and the effects of root alkaloids on the proliferation of two kinds of tongue cancer cells were detected by MTT assay. The effect of different concentrations of root alkaloids on apoptosis was detected by flow cytometry (Annexin V-FITC / Pi staining). Western blotting was used to detect the effect of different concentrations of root alkaloids on the expression of NF- 魏 B in cells. The effect of different concentrations of root alkaloids on the migration ability of tongue cancer cells was detected by cell scratch assay. Results: the proliferation of tongue cancer cell lines Tscca and Tca8113 treated by root alkaloids was significantly inhibited. IC50 was 2.53 渭 mol / L / L 2.38 渭 mol / L 1.96 渭 mol / L for 24 h / 48 h / 72 h, respectively. And there was a dose-dependent relationship between them. The migration of Tca8113 cell line was inhibited by a certain concentration of root alkaloid. Conclusion: root alkaloid can significantly inhibit the proliferation and induce apoptosis of Tscca and Tca8113 in tongue cancer cells, and its mechanism may be related to the effect on the expression of NF- 魏 B. At the same time, root alkaloids could affect the migration of Tca8113 cell lines.
