脱氧紫色杆菌素抑制结肠癌TH29细胞增殖及诱导凋亡作用与机制

【摘要】:目的探讨脱氧紫色杆菌素(Deoxyviolacein)对人结肠癌HT29细胞增殖抑制和诱导凋亡及其可能机制。方法用不同浓度的脱氧紫色杆菌素处理HT29细胞,采用四甲基偶氮唑蓝(MTT)比色法,检测脱氧紫色杆菌素对HT29细胞的抑制率;流式细胞术检测细胞周期和凋亡;Western Blot法检测凋亡相关蛋白Bax、Bcl-2和Caspase9表达的变化。结果脱氧紫色杆菌素对HT29细胞有明显的增殖抑制作用;可提高G0/G1期细胞比例,降低S期细胞比例;细胞凋亡率升高,Bax和Caspase9蛋白表达升高,Bcl-2蛋白的表达降低。结论脱氧紫色杆菌素抑制结肠癌HT29细胞增殖并诱导凋亡,其凋亡机制可能作用于线粒体介导的内源性通路。
[Abstract]:Objective to investigate the inhibitory effect of deoxyviolacein on the proliferation and apoptosis of human colon cancer HT29 cells and its possible mechanism. Methods HT29 cells were treated with different concentrations of deoxyviolacein. The inhibitory rate of deoxypurple bactein on HT29 cells, the cell cycle by flow cytometry and the expression of apoptosis-related protein BaxanBcl-2 and Caspase9 were detected by flow cytometry. Results Deoxypurple bactein could inhibit the proliferation of HT29 cells. It can increase the proportion of G _ 0 / G _ 1 phase cells and decrease S phase cell ratio, increase the expression of Bax and Caspase9 protein and decrease the expression of Bcl-2 protein. Conclusion Deoxypurple bactein can inhibit the proliferation of colon cancer HT29 cells and induce apoptosis. The mechanism of apoptosis may play an important role in mitochondrial mediated endogenous pathway.
